Insights into the Protonation State and Counterion Interactions of Linear Polyethyleneimine Materials.

Byrne Agustín J AJ, Vladimir Daniel D, Garro-Linck Yamila Y, Copello Guillermo J GJ et al.

Polyethyleneimine (PEI) polymers are widely used in the synthesis of nanoparticles, hydrogels and composite materials for biotechnological and environmental applications. However, the solid-state structural organization of linear PEI (L-PEI) materials, and how it is influenced by molecular weight, protonation state and counterion type, have not yet been comprehensively studied. In this work, solid-state nuclear magnetic resonance (ss-NMR), X-ray powder diffraction (XRD), small-angle X-ray scattering (SAXS) and time-domain NMR (TD-NMR) experiments were combined with theoretical calculations to analyze the local chemical environments, crystalline and nanoscale organization, and molecular mobility of L-PEI materials with different molecular weights. The analyzed materials were the free base (PEI·FB), the hydrochloride (PEI·HCl), the trifluoroacetate (PEI·TFA), and the sulfate-containing forms of L-PEI (PEI·Na2SO4 and PEI·H2SO4), as well as an intermediate protonation state obtained by the heterogeneous treatment with HCl-saturated isopropanol. Proton, 13C or 15N ss-NMR studies evidenced different chemical environments associated with changes in protonation state and counterion identity. Furthermore, XRD and SAXS showed that these variables, together with molecular weight and sample preparation conditions, influenced the crystalline arrangement and characteristic nanoscale organization of the polymer chains. Moreover, TD-NMR revealed the presence of three dynamically distinct proton populations, assigned to rigid, interfacial, and mobile domains, whose relative contributions and relaxation behavior varied among the PEI materials. Finally, high-resolution magic-angle spinning (HRMAS) NMR experiments were used to study different PEI-gel materials and films as well as the crosslinking of protonated PEI and sulfate ions.

Dual-Drug Chitosan Hydrogel Loaded with Metformin and Cefazolin for Antibacterial, Anti-Inflammatory and Antioxidant Treatment of Drug-Resistant Bacteria-Infected Skin Wounds.

Zhang Yilin Y, Cao Jie J, Yan Yong Y, Li Zhaoyang Z et al.

Bacterial-infected skin wounds, especially those caused by drug-resistant pathogens accompanied by persistent inflammation, remain a major clinical therapeutic challenge. Rising antimicrobial resistance compromises the therapeutic effect of conventional single-agent antibiotics, creating an urgent demand for novel wound therapeutics that integrate bactericidal potency, immunomodulation and tissue regeneration capacity. In this work, an injectable multifunctional chitosan-based hydrogel (C@MC) was developed, which simultaneously incorporates metformin hydrochloride (as a potential antibiotic adjuvant) and cefazolin sodium (as a β-lactam class antibacterial drug). The optimized C@MC possesses interconnected three-dimensional pores with an initial storage modulus of ~0.83 kPa (C@MC360), featuring desirable mechanical strength, injectability, tissue adhesion and dual pH/glucose-responsive sustained drug release. In vitro evaluations revealed the dose-dependent bactericidal activity of C@MC against Staphylococcus aureus (S. aureus), Escherichia coli (E. coli) and New Delhi metallo-β-lactamase-1-producing E. coli (NDM-1 E. coli, resistant bacteria). The highest drug-laden group nearly eliminated all wound bacteria in vitro. It scavenged reactive oxygen species (ROS) to lower cellular MDA levels and suppress pro-inflammatory TNF-α and IL-6 in LPS-stimulated RAW 264.7 macrophages. In full-thickness mouse wounds infected with NDM-1 E. coli, C@MC360 achieved nearly complete wound closure, with only a 4.2% residual wound area on day 14, accompanied by suppressed inflammatory infiltration and accelerated wound regeneration. This dual-drug hydrogel integrates antibiotic potentiation, anti-inflammation and antioxidation, providing an intervention regimen for skin wounds infected with drug-resistant bacteria.

Effects of microencapsulated ε-Polylysine hydrochloride on quality and microbial indicators of salted bamboo shoots.

Cai Xianfen X, Du Haonan H, Wang Yang Y, Fei Sitao S et al.

Salted bamboo shoots are susceptible to microbial spoilage and quality deterioration, while ε-polylysine hydrochloride (ε-PL·HCl) suffers from poor stability and hygroscopicity. To address this, ε-PL·HCl-loaded microcapsules were fabricated via complex coacervation using soy protein isolate (SPI) and chitosan (CS) as wall materials. The resulting microcapsules achieved an encapsulation efficiency of 98.63% and exhibited enhanced thermal stability, with a 25-30 °C increase in decomposition temperature compared with free ε-PL·HCl. SEM and XRD supported the incorporation of ε-PL·HCl into the SPI-CS matrix, while in-vitro release studies demonstrated sustained-release behavior in an aqueous medium. During 13 days of storage of salted bamboo shoots, ε-PL·HCl microcapsules (ε-PL·HCl-MCs) maintained hardness at approximately 300 N, reduced total bacterial counts to below 1 × 107 CFU/g, delayed the decrease in b* value (yellowness), and slowed the decline in pH. This SPI-CS-based delivery system provides a promising strategy for natural antimicrobial preservation.

Interactions of Veterinary Antibiotics with Purified Bovine Milk Alkaline Phosphatase: A Combined In Vitro Kinetic and Theoretical DFT/HF/M06-2X Study.

Bilen Çiğdem Ç, Tüzün Burak B, Gencer Nahit N

Alkaline phosphatase (AP, EC 3.1.3.1) is a key metalloenzyme and has been widely used as a reliable marker for milk pasteurization. Milk containing antibiotic residues tends to inhibit AP function, which can result in faulty pasteurization determination. AP from bovine milk was purified by a purification strategy involving butanol homogenization, rennet precipitation, and Sephadex G-100 gel filtration chromatography. The enzyme was purified 53.43-fold with a specific activity of 7516.40 EU/mg. Kinetic characterization showed a KM value of 2.27 mM (Vmax value: 555.56 EU/min); substrate was p-nitrophenyl phosphate. Electrophoretic studies verified the purity and molecular nature of the product enzyme. To functionally validate the identity of the purified enzyme, tetramisole hydrochloride was evaluated as a standard reference inhibitor, yielding a potent IC₅₀ value of 0.045 mM. Fifteen veterinary drugs and antibiotics were used to test the inhibitory effects of purified milk AP in vitro. Among the antibiotic compounds tested, Danofloxacin Mesylate induced the strongest antibiotic inhibition (IC50: 3.36 mM). Gentamycin Sulfate, Metamizole Sodium, Enrofloxacin, Kanamycin Sulfate, Sulfadimidine Sodium, and Neomycin Sulfate had IC50 values ranging between 22.89 and 312.13 mM of inhibition in AP, while Dexamethasone, Oxytetracycline Hydrochloride, Marbofloxacin, Tilmicosin, Doxycycline Monohydrate, Tylosin, Florfenicol and Flunixin Meglumine made no detected inhibition in the presence of 0.5 mM p-nitrophenyl phosphate. The molecular docking studies were additionally performed that the density functional theory (DFT), Hartree-Fock (HF), and M06-2X calculations were used to validate the experimental results with the 6-31++G(d,p) basis set. The theoretical results also reported that Oxytetracycline Hydrochloride, Tilmicosin, and Doxycycline Monohydrate displayed superior electronic reactivity, lower energy gaps between HOMO-LUMO, and more electrophilic character compared with the other compounds. These results are the first complete assessment of the impact of veterinary antibiotics in milk AP and underscore the possible role of antibiotic residues in establishing the reliability of pasteurization control measures.

Structure-Based Design and Biological Evaluation of Imidazo[1,2-a]pyridine Derivatives as Potential Antituberculosis Agents.

Chaugule Hanmant N HN, Jadhav Shravan Y SY, Ghorpade Sujay A SA, Kore Dayanand C DC et al.

A library of 11 novel imidazopyridine-based sulfonamide derivatives (4a-4k) were synthesized through the coupling of an imidazo[1,2-a]pyridine hydrochloride intermediate with various substituted sulfonyl chloride derivatives (3a-3k). The structures of the compounds (4a-4k) were confirmed using appropriate spectroscopic techniques. The synthesized derivatives (4a-4k) were subsequently evaluated for their antitubercular and antioxidant potential. Among the tested molecules, compounds 4b, 4d, and 4g demonstrated moderate antimycobacterial activity, displaying minimum inhibitory concentration (MIC) values of 6.21 ± 0.45 µg/mL, which were comparable to that of the standard drug rifampicin. To gain insight into their possible mechanism of action, molecular docking studies were carried out against selected tuberculosis (TB)-associated protein targets. The active compounds exhibited favorable binding interactions within the target binding pockets, supporting the observed biological activity. Furthermore, in silico ADMET assessment revealed encouraging pharmacokinetic characteristics, including good gastrointestinal absorption, satisfactory bioavailability, and compliance with common drug-likeness criteria without significant rule violations. Collectively, these findings identify the imidazopyridine sulfonamide derivatives as potential lead structures for further structural optimization and biological investigation toward the development of new antitubercular agents.
