A comprehensive characterization strategy incorporating multiple filtration approaches and enhanced isomer identification: A case study of Eleutherococcus senticosus leaves.

Eleutherococcus senticosus (Rupr. & Maxim.) Maxim leaves (ESL) are consumed as tea or stir-fried dishes and exhibit neuroprotective activity. Their primary active components, triterpenoids, pose characterization challenges due to structural complexity and extensive isomerism. To address these challenges, we developed a novel integrated annotation strategy that incorporates polygonal mass defect filtering (p-MDF), molecular fingerprinting, molecular networks (MN) and ion mobility spectrometry (IMS). The EstMIL multidimensional database was established, encompassing 305 triterpenoids and addressing the challenge of insufficient database coverage. Finally, the integration of collision cross-section (CCS) with FBMN enhanced the characterization of unknown chemical compositions and improved the ability to identify isomers. This approach identified 13 triterpenoid isomer pairs, 6 of which were validated by ER-MS and quantum chemical calculations, confirming the reliability of the strategy. This integrated strategy ultimately identified 203 triterpenoids, comprising 64 newly identified compounds and 48 with novel potential.

Backbone-Mediated Coordination-Environment Reshaping in Gel Polymer Electrolytes for High-Voltage Lithium Metal Batteries.

Conventional linear polyether electrolytes feature abundant ether-oxygen sites that dominate the primary Li+ solvation shell, limiting anion participation and undermining ion transport and high-voltage stability. A backbone-mediated coordination-environment reshaping strategy is proposed by incorporating diglycidyl 1,2-cyclohexanedicarboxylate (DCD) into the copolymerization of 1,3-dioxolane (DOL), yielding an in situ crosslinked gel polymer electrolyte, P(DCD-DOL). The weakly coordinating ester carbonyl groups and the spatially restricted ether-oxygen sites in the crosslinked network jointly weaken backbone-Li+ coordination and promote anion/solvent participation in the Li+ solvation shell. Multiscale characterizations and theoretical simulations confirm that P(DCD-DOL) effectively optimizes the Li+ solvation structure, leading to a high Li+ transference number of 0.64, an oxidative stability window of 5.20 V, and inorganic-rich electrode-electrolyte interphases. Consequently, Li||NCM811 cells with P(DCD-DOL) retain 84.21% capacity after 500 cycles at 4.3 V, and 80.62% after 200 cycles at 4.5 V. Moreover, a 0.9 Ah pouch cell achieves an energy density of 305 Wh kg-1 and retains 82.33% capacity over 180 cycles. This work highlights polymer backbone engineering as an effective route to regulate Li+ coordination and interfacial chemistry in high-voltage lithium metal batteries.

Advancing genetic evaluation of milk yield and composition using a genomic-polygenic model in smallholder dairy cattle farms in Thailand.

Improving the accuracy of genetic evaluation in smallholder dairy systems is essential for sustainable productivity. However, traditional polygenic models (PM) are often constrained by incomplete pedigree, heterogeneous management, and limited genotyping resources. This study evaluated a genomic-polygenic model (GPM) relative to a PM using data from a multibreed dairy population raised under Thai tropical smallholder conditions. Phenotypic records for 305-day milk yield (MY), fat percentage (FP), protein percentage (PP), and somatic cell count (SCC) from 14,417 first-lactation cows across 1,321 farms were analyzed together with genotypes from 5,479 animals generated using GeneSeek Genomic Profiler (GGP) arrays ranging from 9 K to 150 K SNPs. Both models included herd-year-season of calving, age at first calving, and heterosis as fixed effects, and additive genetic and residual components treated as random. The GPM yielded higher additive genetic variances and heritability estimates and produced more biologically consistent antagonistic correlations among traits than the PM. Prediction accuracy was improved for all animal groups under the GPM, with the largest gain observed in genotyped young sires (18.36%). Pedigree connectedness analysis indicated that genotyping animals with low to moderate relationships enhances accuracy cost-effectively. Despite persistent challenges associated with heterogeneous management and limited pedigree depth, the results demonstrate the practical value of genomic-polygenic evaluation for smallholder multibreed dairy populations in tropical environments.

Changes in Alcohol Use Among Adults with Cancer Participating in the ECOG-ACRIN EAQ171CD Tobacco Treatment Trial.

Alcohol and tobacco use are known carcinogens. Treating alcohol and tobacco co-use can improve treatment and survivorship outcomes among patients diagnosed with cancer. We conducted a post hoc analysis of ECOG-ACRIN EAQ171CD trial participants within the NCI Community Oncology Research Program (NCORP), examining changes in alcohol use among adults receiving the virtual sustained treatment (VST) for tobacco use intervention versus enhanced usual care (EUC; Quitline referral). Multivariable logistic regression assessed changes in alcohol use disorder risk at 3 and 6 months using the AUDIT-C. At baseline, participants (N = 305) were classified as no risk (31%), low/minimal risk (38%), moderate risk (23%), or high risk (8%) for alcohol use disorder. The mean age was 55.6 years, participants smoked for an average of 33 years, and 18% had alcohol-associated tumors. Participants randomized to EUC were more likely to shift to a lower alcohol risk category than those in the VST arm at 3 months (39% vs 27%) and 6 months (33% vs 27%). There was no significant difference in the odds of shifting to a lower risk category for VST vs EUC at 3 months (adjusted OR = 0.56, 95% CI 0.26-1.18, p = 0.125) or at 6 months (adjusted OR = 0.57, 95% CI 0.25-1.28, p = 0.172). Patients diagnosed with cancer and engaging in alcohol and tobacco co-use can achieve reductions in alcohol use and risk. Integrating tailored, co-use-focused strategies into cessation programs may improve changes in modifiable risk behaviors.

Prevalence of blood-borne viral infections and HBV vaccination coverage among people with recent injecting drug use in Oslo, 2002-2024.

We assessed changes in the prevalence of HCV, HBV, HIV and HBV vaccination coverage among people with recent injecting drug use in Oslo between 2002 and 2024. We used data from eight cross-sectional studies among PWID in Oslo in 2002, 2012, 2015, 2018 and 2021-2024. Participants (≥18 years, injected drugs within the past four weeks) completed venipuncture and a questionnaire. Outcomes were detectable HCV RNA, HIV Ag/Ab, HBsAg, and HBV vaccination coverage, defined as anti-HBs ≥10 IU/L among anti-HBc negative individuals. Factors were analysed using generalised estimating equations. We included 1689 participants. HCV RNA prevalence decreased from 50.2% (95% CI 44.4-55.9) in 2002 (153/305) to 6.3% (95% CI 3.5-10.4) in 2024 (14/221). Detectable HCV RNA was associated with year of participation (2024 vs 2002; aOR 0.06; 95% CI 0.03-0.11), age (aOR 1.30 per 10-year increment; 95% CI 1.14-1.49) and daily injecting drug use (aOR 1.33; 95% CI 1.01-1.74). The prevalence of HBsAg and HIV remained low (<3.3%) throughout. In 2021-2024, 40.0% (95% CI 35.3-44.8; 170/425) of those not exposed to HBV were vaccinated. HBV vaccination was associated with recent use of a drug consumption room (aOR 1.33; 95% CI 1.02-1.72). HCV RNA prevalence among people with recent injecting drug use in Oslo decreased significantly between 2002 and 2024, while the prevalence of HIV and HBsAg remained low and stable. Future efforts should focus on identifying and treating the remaining population infected with HCV, increasing HBV vaccination uptake, and maintaining low HIV prevalence.

Long-term effects of dairy calf social housing on first lactation survival and milk yield.

Given advantages of preweaning social housing for dairy calf performance and behavioral development, the objective of this study was to characterize long-term effects of social housing on survival and milk yield during first lactation. Holstein heifers were previously raised in individual housing (IH; n = 90), pair housing (PH; n = 60 pens, 120 heifers), or in groups of 10 (GH; n = 12 pens, 80 heifers) for the first 2 mo of life, then commingled among housing treatments and raised in groups on pasture where they underwent standard rearing and breeding practices. At 30 d before expected calving, heifers were moved to a free stall barn. Following calving, heifers entered the lactating herd where they were milked twice daily and followed until the completion of their first lactation. We found no effect of social housing on pre-calving culling risk, but socially housed heifers were less likely to be culled during lactation (PH: hazard ratio = 0.35, 95% CI: 0.16-0.77, and GH: hazard ratio = 0.48, 95% CI: 0.25, 0.92). In cows that completed a 305 d lactation, social housing also resulted in greater first lactation milk yield, with greatest effects in the first 100 d post-calving. Effects of treatment on milk yield were predominantly direct, with pre-weaning ADG having a partial mediating effect explaining 15% of the total treatment effect. Early lactation milk yield, estimated in wk 2 to 8 for all cows regardless of culling events in lactation, predicted lower culling risk in PH and GH, compared with IH, suggesting that reduced milk yield over the full lactation in IH animals was exacerbated due to involuntary culling. These results support long-term production advantages attributed to social housing and provide some evidence to suggest that long-term effects of social housing may be attributed to behavioral mechanisms resulting in increased resilience rather than differences in biological potential arising from pre-weaning growth.
