Antioxidant Activities of Pentacyclic Triterpenes From Melipona beecheii Propolis and Their Semisynthetic Derivatives.

Four principal fractions (I-IV) were obtained from the Melipona beecheii propolis. GC-MS analysis of these fractions allowed us to recognize 11 pentacyclic triterpenes (PTs), fraction I [β-amyrin acetate (1), germanicol acetate (2), moretenol acetate (3) and 24-methylencycloartan-3-ol acetate (4)]; fraction II [marsformosanone (5), β-amyrenone (6), germanicone (7) and lupenone (8)]; fraction III [β-amyrin (9) and, glutinol (10)]; and fraction IV [9 and α-amyrin (11)]. This is the first time that triterpenes 7 and 10 are reported in the propolis of M. beecheii. Three semisynthetic derivatives were prepared: the reduction of fraction II and the acetylation and oxidation of fraction IV. The ethanolic extract of propolis and fraction III presented the highest free DPPH radical scavenging capacity at 53.89 ± 8.39% and 47.77 ± 0.91%, respectively, using a concentration of 10 mg/mL. In contrast, the reducing power of Fe (III) analysis showed a strong reducing power of Fe(III) for all fractions and their derivatives with an EC50 = 0.8-1.0 mg/mL.

Transient infrared laser exposure modulates calcium activity in cortical dendritic spines.

Infrared neural stimulation (INS) is an optical neuromodulation technique that elicits neural activity through photothermal temperature gradients. Photothermal gradients have been shown to alter neuronal excitability, but the effects on the dendritic spine (DS) structure and function have not been characterized. We aim to determine whether INS induces calcium signaling in DS and whether this signaling alters filamentous actin (F-actin). We used confocal imaging of cortical neurons expressing a genetically encoded calcium indicator (GCaMP)8f-Syn1 and mCardinal-LifeAct during laser exposure. Fluorescence signals were measured within regions of interest covering the DS. The resulting fluorescence measurements were baseline-normalized, and transient spiking activity in DS was quantified. Transient thermal gradients induce persistent calcium activity in a fraction of DS, independent of pulse duration but dependent on the absolute temperature and number of laser exposures. There is no relationship between the calcium activity and F-actin. Calcium signaling in DS was not mediated by ionotropic glutamate receptors but depended on extracellular entry. These results indicate that transient thermal gradients elicit calcium activity in DS, but the induced activity does not alter F-actin, suggesting that transient thermal gradients may be a useful tool for studying calcium dynamics in DS without triggering traditional structural plasticity pathways.

Direct targeting of ORAI1 by ginsenoside Rg3 modulates calcium signaling and senescence-associated AMPK-NRF2 activation.

20(S)-ginsenoside Rg3 (Rg3(S)), a major saponin derived from red ginseng, exhibits diverse biological activities, including antioxidant and anti-senescence effects. However, the direct molecular targets through which Rg3(S) regulates calcium signaling and its role at membrane contact sites during cellular senescence remain largely unknown. Plasma membrane (PM)-endoplasmic reticulum (ER) contacts and related protein interactions were analyzed using proximity ligation assays and co-immunoprecipitation. Direct binding of Rg3(S) to ORAI1 was validated using cellular thermal shift assays and microscale thermophoresis. Molecular docking simulations followed by site-directed mutagenesis were used to define critical binding residues. Cytosolic calcium levels and cellular senescence were assessed using calcium imaging and senescence-associated β-galactosidase staining. Rg3(S) increased cytosolic calcium levels independently of ER calcium depletion and was accompanied by a reduction in PM-ER contacts. Rg3(S) directly bound to ORAI1 in a dose-dependent manner, identifying ORAI1 as a previously unrecognized molecular target of ginsenoside Rg3. Molecular docking revealed LYS204 and ILE229 within the extracellular loop of ORAI1 as key residues maintaining this interaction. Mutation of these residues abolished Rg3(S)-induced calcium influx, leading to impaired activation of the AMPK-NRF2 pathway and attenuation of the anti-senescence effect of Rg3(S). These findings identify ORAI1 as a key molecular mediator of ginsenoside Rg3(S)-induced calcium signaling linked to cellular senescence. By modulating PM-ER contact sites and cytosolic calcium dynamics, Rg3(S) attenuates senescence, providing new mechanistic insight into the anti-aging potential of ginseng-derived compounds beyond autophagy-centered pathways.

Manipulating the Buried Interfacial Dipole: The Role of Pyridine Acetate-Hydrochloride Isomers in Carbon-Based Perovskite Solar Cells Prepared in Air.

The buried interface between tin oxide and perovskite is the key factor for non-radiative recombination and energy level mismatch, which limits the performance and stability of perovskite solar cells. This work explores a simple interfacial dipole engineering strategy, where three pyridine acetate-hydrochloride (PAH) isomer molecules (2-PAH, 3-PAH, and 4-PAH) are used to modify the SnO2 electron transport layer. The 3-PAH-modified layer can control the work function of tin oxide, achieve the best energy level alignment, and improve the crystallization quality of the perovskite film, thereby effectively suppressing interface recombination and promoting electron extraction. All the devices are prepared in air, and the device optimized by 3-PAH achieved a champion energy conversion efficiency of 14.31% and demonstrated stability. Subsequent to an 800 h placement in an N2 glove box or a 340 h exposure to an air environment, the unencapsulated target devices that were modified by 3-PAH maintained 80.7% and 81.3% of their initial efficiency.

The evolution of Ca2+-ATPases across plants with profiles in Rhododendron and the function of key members in alleviating high calcium stress.

Ca2+-ATPase (CAP) is a key Ca2+ efflux protein in plants. Our previous research suggests that CAPs may play a crucial role in the adaptation of rhododendrons to high calcium environments. However, the evolution, variation, characteristic expression, and subfunctionalization of this gene family in Rhododendron remain unknown. Through the analysis of pan-genomes and pan-transcriptomes, we elucidated the systematic evolution of CAPs in plants, as well as their characteristic expression patterns in Rhododendron. During the evolutionary process from lower to higher plants, CAPs can be divided into six clades and exhibit structural conservation. CAPs have emerged and differentiated in lower plants such as algae, and they have undergone significant amplification in Eudicots plants like rhododendrons. Three Rhododendron species (Rhododendron bailiense, R. delavayi, and R. irroratum) located in the karst province of Guizhou in Southwest China exhibit the highest copies of CAPs, suggesting a strong association between CAP copy number variation and habitat, particularly in high calcium environments. Through multiple transcriptome analyses, we revealed that CAPs are induced under various environmental/developmental conditions (e.g. karst environments, high altitude, early flower development, hormones, etc.). Co-expression network analysis highlighted key members of calcineurin B-like protein (CBL) and CBL-interacting protein kinases (CIPK) that are associated with the high expression of CAPs. Experimental validation demonstrated that CAPb1 and CAPd1 significantly alleviate high calcium stress, and the CAPb1-CIPK1-CBL1 and CAPd1-CIPK2-CBL1 modules can further enhance the alleviation. These findings provide new insights into the evolution, characteristic expression, and function of CAPs, as well as new perspectives on the high calcium adaptability of rhododendrons.
