Adaptive Laboratory Evolution Rewires Pseudomonas putida for Resource-Efficient Acetate Assimilation.

Gurdo Nicolás N, Srinivasan Aparajitha A, Tagliani Tommaso T, Filbig Melanie M et al.

Acetate is an attractive renewable two-carbon substrate for microbial biotechnology, but toxicity limits growth and carbon-use efficiency at process-relevant concentrations. Here, we used adaptive laboratory evolution to improve acetate tolerance in a genome-reduced strain of Pseudomonas putida and combined whole-genome sequencing, quantitative physiology, reverse engineering, transcriptomics, proteomics and 13C-acetate fluxomics to resolve the underlying adaptation mechanisms. Evolution under increasing acetate concentrations selected recurrent mutations in gacA and fabB, which encode a global response regulator and a fatty acid biosynthesis enzyme, respectively. Reverse engineering of these mutations recapitulated most of the evolved phenotype, including shorter lag phase and substantially higher biomass yield from acetate. Multi-omic analyses showed repression of type VI secretion systems, carbohydrate storage functions, fatty acid metabolism and oxidative stress-associated proteins, indicating resource reallocation away from costly stress and non-essential programs. Fluxomics further revealed reduced carbon cycling and increased glyoxylate shunt flux, consistent with improved acetate-carbon retention in biomass. These results establish acetate tolerance in P. putida as a resource-efficiency phenotype and identify gacA and fabB as actionable targets for acetate-based bioproduction.

Multi-Electron Transfer in Zinc-Based Batteries: Unraveling Redox Mechanisms Toward Next-Generation Self-Powered Technologies.

Zhou Chao C, Zhou Jiaju J, Guo Xiaojun X, Zhang Haoxiang H et al.

The rapid expansion of autonomous IoT networks and wearable electronics has intensified the demand for photo-rechargeable zinc-based batteries (PRZBBs). While realizing high overall energy conversion efficiency intrinsically relies on cathode materials capable of multi-electron transfer reactions, current literature largely treats the fundamental electrochemistry of aqueous zinc batteries and the macroscopic engineering of solar-integrated devices as isolated domains. This review bridges this critical gap, offering a unique positioning by establishing a unified analytical framework that directly links fundamental multi-electron redox mechanisms to the dynamic architectural requirements of PRZBBs. Moving beyond a conventional cataloging of material classes, we critically evaluate diverse cathode platforms, including transition metal oxides, Prussian blue analogues, organics, halogens, and sulfur. Our incremental contribution lies in decoupling how specific multi-step reaction kinetics, distinct voltage plateaus, and dominant charge carriers dictate solar cell coupling strategies, particularly concerning maximum power point matching and intermittent charging adaptability. Furthermore, we critically and systematically assess the practical bottlenecks of these chemistries, such as shuttle effects, kinetic limitations, and voltage hysteresis, that hinder scalable integration. Finally, we distill these insights into a prioritized, roadmap-oriented perspective, outlining the short- to long-term engineering and scientific milestones necessary to translate fundamental multi-electron zinc chemistry into next-generation, commercially viable self-powered technologies.

SpectralMol: Spectral Graph Bisection Achieves Universal Molecular Fragment Decomposition for Drug-Like Molecule Generation.

Li Gang G, Lin Zhizhe Z, Zhou Teng T, Bai Weihua W et al.

Generating valid, diverse, and drug-like molecules remains a central challenge in computational drug discovery. A key bottleneck for fragment-based generative models is decomposition coverage: existing junction-tree methods fail on bridged bicyclic and spiro ring systems common in drug-like scaffolds. Our primary contribution is Fiedler-BFS (breadth-first search) molecular decomposition, a spectral bisection algorithm achieving 100% decomposition success on all 250,000 ZINC-250K molecules, including bridged and spiro systems where junction-tree methods fail. A decomposition ablation (5,000 molecules, 5 seeds) confirms that spectral structure drives the improvement: 91.4% vs 7.1% for random bisection. Compared to BRICS (break retrosynthetically interesting chemical substructures; a rule-based retrosynthesis method), Fiedler-BFS produces fewer fragments per molecule (2.31 vs 5.13), better suited for hierarchical generative modeling. As a secondary contribution, we instantiate the decomposition in a Tree-VAE (variational autoencoder) with classifier-free guidance over molecular properties, Inverse Multi-Quadratic MMD (maximum mean discrepancy) regularization, and gradient-based latent-space steering. Under a matched protocol in which the identical Tree-VAE is retrained while only the cut-bond set varies, Fiedler-BFS yields shorter, more learnable fragment trees than BRICS and junction-tree decomposition (validation reconstruction loss of 0.012 vs 0.048 vs 0.083). On ZINC-250K, the model achieves 100% chemical validity, 99.87% novelty, a mean QED (quantitative estimate of drug-likeness) of 0.603 (matching the ZINC-250K training mean), and an SA (synthetic accessibility score) of 2.70, confirming compatibility with a hierarchical generative pipeline for drug-like molecule generation. Property conditioning reaches only a 12.2% QED hit rate and, under robust metrics, does not outperform an unconditioned baseline; the v2 vocabulary naturally generates drug-like molecules with QED ≈ 0.60, so stronger property guidance remains an open challenge.

Engineering the Catalytic Sites and Porous Framework of Biomass-Derived Carbon Aerogels for Zinc-Air Batteries and Pressure Sensors: A Review.

Yu Chenyue C, Chen Yanglei Y, Zhang Lei L, Li Ru R et al.

The ever-growing global demand for sustainable energy has promoted the development of zinc-air batteries with high energy density. However, the slow multi-electron kinetic process of the oxygen reduction reaction and oxygen evolution reaction still restricts the practical commercialization of this battery technology. Biomass-based carbon aerogels can be constructed into tunable three-dimensional hierarchical structural frameworks, which provide high specific surface area and tunable structural characteristics, offering opportunities to improve mass transport and catalytic-site accessibility. This paper systematically reviews the latest research progress in the preparation and structural regulation of biomass-based carbon aerogels and elaborates on the determining effect of directional sol-gel processes, advanced drying techniques, and activation mechanisms on pore structure. To further improve the bifunctional electrocatalytic activity, this paper highlights the key doping and modification strategies, including nitrogen doping, multi-element co-doping, as well as the precise immobilization of non-noble metal nanoparticles and single-atom active sites. In addition to serving as high-efficiency, self-supporting, and flexible air cathodes for energy storage devices, the excellent mechanical rebound performance and continuous conductive network of this type of carbon aerogel also make it suitable for high-sensitivity pressure sensors. In conclusion, rational design and modification of biomass precursors can open up a green and sustainable development path for next-generation multifunctional flexible electronic devices.

Morphological, anatomical, and phytochemical characterization of Sisymbrium altissimum L.: chemical composition, biological activities, and isolation of major constituents.

Tekman Enes E, Aydın Bilge B, Yuca Hafize H, Özbek Hilal H et al.

This study comprehensively evaluated the phytochemical composition and multidimensional biological activities of Sisymbrium altissimum L., a wild edible species belonging to the Brassicaceae family. Methanolic extracts obtained from aerial parts, flowers, fruits, and roots were successively fractionated using solvents of increasing polarity, and essential and fatty oils were characterized by GC-MS and GC-FID analyses. The essential oil composition showed marked organ-dependent variation, with methallyl cyanide dominating fruit (91.9%) and flower (75.9%) oils, while phenylethyl isothiocyanate (48.7%) was the major constituent in roots. Further phytochemical investigation of the biologically active root hexane extract led to the isolation of β-sitosterol, lauric acid, and β-sitosterol-3-O-glucoside. Biological activity assays revealed strong α-glucosidase inhibition, particularly in the root hexane extract (87.92%), exceeding the reference drug acarbose (68.49%), whereas α-amylase inhibition was moderate, with the root ethyl acetate fraction exhibiting the highest activity (40.84%). Cholinesterase inhibition assays demonstrated low to moderate acetylcholinesterase and butyrylcholinesterase inhibitory activities depending on the plant part and extract fraction. Hyaluronidase inhibition was notably high, reaching up to 98.70%, whereas moderate collagenase (27.56%) and elastase (23.98%) inhibition and low tyrosinase activity were observed. Antioxidant activity was prominent in ethyl acetate and n-butanol fractions, with ABTS radical scavenging values approaching those of standard antioxidants. Total phenolic and flavonoid contents correlated with antioxidant capacity, although structural factors also influenced activity. Importantly, all tested extracts and cold-pressed oil exhibited no mutagenic or genotoxic effects in the Ames/Salmonella assay. Overall, these findings provide a comprehensive characterization of S. altissimum and highlight its potential as a source of biologically active natural products.

Structural evolution-induced strong-coupling in CuRu-Cu2O nanocomposite enables low-overpotential electrocatalytic ammonia synthesis.

Zhang Ying Y, Zhang Tong T, Gao Tianyi T, Lv Jiangnan J et al.

Ammonia (NH3) synthesis via nitrate reduction offers a sustainable energy conversion and mitigates aquatic NO3- pollution, yet most catalysts require high overpotentials for appreciable NH3 yields. Here, we report a sulfur-leaching strategy to transform Cu1.8S/Ru heterostructures into a small CuRu-Cu2O nanocomposite with abundant heterointerfaces, enabling strong electronic coupling between CuRu and Cu2O particles, synergistically enhancing NO3- adsorption and reaction kinetics. The optimized catalyst achieves 95.4% NH3 Faradaic efficiency at a low potential of 0.0 V versus reversible hydrogen electrode, stable over 30 cycles. Operando spectroscopy and density functional theory calculations reveal the incorporation of Ru facilitates active hydrogen generation, alleviating the energy barrier for hydrogenation steps at heterointerfaces. The catalyst demonstrates high stability in a membrane electrode assembly at 450 mA cm-2 over 200 h and enables a Zinc-NO3- battery operation exceeding 50 h, offering a design model for durable, active nanocomposites in sustainable NH3 synthesis.
