Hypoxia-Responsive Fluorescent Prodrugs: Self-Reporting Drug Delivery or Activation in Living Systems.

Hu Xiaomu X, Song Xiaoyu X, Qiu Yijie Y, Du Wei W

Hypoxia is a hallmark of numerous pathological conditions, such as cancer, drug-induced liver injury, and rheumatoid arthritis. Given the contrast in oxygen levels between pathological and normal tissues, hypoxia-activated prodrugs have been developed by incorporating hypoxia-responsive groups to achieve targeted therapy while minimizing toxicity to normal cells. Recently, the integration of drug delivery or activation (including triggered release) with fluorescence imaging has emerged as a promising strategy in prodrug design, enabling self-reporting of drug processes and facilitating preclinical evaluation of prodrug performance. This review provides a comprehensive overview of hypoxia-responsive fluorescent prodrugs capable of self-reporting drug delivery or activation/release via fluorescence readouts. We summarize recent advancements in such prodrugs, focusing on design strategies, drug release or activation mechanisms, fluorescence signal modulation, and biomedical applications, especially in cancer treatment. Furthermore, we discuss current challenges and future perspectives for hypoxia-responsive fluorescent prodrugs, as well as potential directions for the development of the next generation of hypoxia-responsive prodrugs.

Vitamin B12-Functionalized Nanogels for pH-, and Redox-Responsive Doxorubicin Delivery.

Dagdelen Serife S, Mestizo Paula D PD, Mackiewicz Marcin M, Kurdyn Agnieszka A et al.

Rapidly proliferating cancer cells display an elevated demand for vitamin B12, rendering this bacterial-derived cofactor an attractive targeting motif for nanotherapeutic drug delivery. Herein, we report a concise two-step synthesis of a polymerizable 5'-modified vitamin B12 -monomer that enables the covalent integration of B12 into pH- and redox-responsive nanogels. The resulting nanogels efficiently encapsulate doxorubicin through electrostatic interactions between the cationic drug and the anionic nanogel network, while enabling stimuli-responsive drug release under tumor-relevant conditions. In vitro studies revealed that DOX@B12-nanogels retain potent antiproliferative activity against HT-29 colorectal cancer cells (IC50 = 0.878 ± 0.102 μM), while decreasing cytotoxicity toward healthy CCD 841 CoN cells by 34-fold relative to non-B12-functionalized control nanogels. These findings establish covalently engineered vitamin B12-nanogels as a promising platform for selective and stimuli-responsive anticancer drug delivery.

Engineering Smart Hydrogels Through Dynamic Polymer Networks for Controlled Drug Delivery.

Choi Chanju C, Seo Dongseong D, Kim Taeho T, Park Jonghyun J et al.

Smart hydrogels are increasingly explored as adaptive platforms for controlled drug delivery because their polymer networks can respond dynamically to chemical, biological, and physical cues. Among these systems, dynamic polymer networks formed through reversible covalent and noncovalent interactions provide a unique means of coupling molecular-scale bond exchange with time-dependent changes in hydrogel structure, mechanics, degradation, and therapeutic transport. This review presents an integrated framework linking dynamic crosslink chemistry, exchange kinetics, network properties, and drug-release behavior. Dynamic covalent, supramolecular, metal-ligand, and hybrid network strategies are compared with particular emphasis on how crosslink lifetime, mesh accessibility, swelling, viscoelasticity, stress relaxation, and degradation govern therapeutic cargo loading, retention, diffusion, and release. Representative release mechanisms and kinetic models are discussed together with cargo-specific design considerations for small molecules, proteins and peptides, nucleic acids, and nanoparticle-based therapeutics. Biomedical applications are further considered according to therapeutic requirements and administration routes, including injectable local depots, topical and transdermal delivery, and regenerative systems. Finally, key translational challenges involving physiological complexity, biocompatibility, reproducibility, scale-up, sterilization, and storage stability are critically assessed. By connecting molecular interaction dynamics with network-level behavior and therapeutic performance, this review provides design principles for developing more predictable, programmable, and clinically translatable dynamic hydrogel drug-delivery systems.

Alpha-mangostin combined with doxorubicin-loaded hydrogel for the treatment of ovarian cancer.

Liu Xia X, Zhou Wenjie W, Chen Lezong L, Wang Yue Y et al.

Ovarian cancer is one of the most prevalent malignancies of the female reproductive system and a gynecological malignancy with a high mortality rate. Alpha-mangostin is a xanthone derivative with anticancer activity that can increase the sensitivity of ovarian cancer cells to doxorubicin, and the combination of the two drugs exerts a synergistic therapeutic effect. However, the poor solubility of α-mangostin limits its application. To solve this problem, we prepared an α-mangostin nanoformulation via self-assembly using the film dispersion method. The formulation had a uniform particle size of approximately 58 nm, a stable nanostructure and good water solubility. Hydrogels, as good thermosensitive drug delivery systems, enable sustained and controlled release of drugs. Accordingly, we utilized the synergistic efficacy of the α-mangostin nanoformulation combined with doxorubicin and used a thermosensitive hydrogel carrier to jointly treat ovarian cancer. In vitro, the combined treatment achieved better effects than either drug alone in inducing ovarian cancer cell apoptosis, inhibiting ovarian cancer cell proliferation, and inducing cell cycle arrest. In vivo, this composite formulation effectively inhibited tumor growth by inducing ovarian cancer cell apoptosis, inhibiting tumor cell proliferation, and suppressing tumor angiogenesis, and effectively inhibited ascites formation, achieving a better antitumor effect than either drug alone. In summary, the composite hydrogel co-loaded with α-mangostin nanoformulation and doxorubicin represents a promising and effective strategy for the treatment of ovarian cancer.

Development and Optimization of a Lupeol-Loaded Bilosomal Gel for Enhanced Topical Therapy of Imiquimod-Induced Psoriasis.

Hamza Mohammad M, Samanta Soumya S, Khan Mohammed Faheem MF, Roy Supriya S et al.

Lupeol, a naturally occurring pentacyclic triterpenoid, exhibits potent anti-inflammatory and antioxidant activities, making it a promising candidate for psoriasis treatment. However, its poor aqueous solubility and limited skin permeation restrict its therapeutic application. This study aimed to develop and optimize a lupeol-loaded bilosomal gel to enhance topical delivery and improve anti-psoriatic efficacy. Lupeol-loaded bilosomes were prepared using the thin-film hydration method and optimized through a Box-Behnken experimental design. The optimized formulation was characterized for vesicle size, entrapment efficiency, drug release, and physicochemical properties before incorporation into a Carbopol® 934P gel. The bilosomal gel was further evaluated through in vitro drug release, ex vivo skin permeation, and in vivo efficacy in an imiquimod-induced psoriasis-like mice model. The optimized bilosomes exhibited nanoscale vesicles, high drug entrapment, and sustained drug release. Incorporation into the gel produced a formulation with suitable physicochemical characteristics and significantly enhanced skin permeation compared with a conventional gel. In vivo, the bilosomal gel markedly reduced Psoriasis Area and Severity Index (PASI) scores, spleen index, and TNF-α levels while restoring normal skin histology. These findings demonstrate that bilosomal encapsulation substantially improves the topical delivery and therapeutic efficacy of lupeol, highlighting the developed bilosomal gel as a promising nanocarrier-based strategy for the topical management of psoriasis.

Temperature-Responsive Antheraea pernyi Silk Fibroin/Poly(N-isopropylacrylamide) Hydrogels for Controlled Protein Drug Delivery.

Wang Boxiang B, Xu Hangdan H, Huang Hong H, Xie Jinyang J et al.

Temperature-responsive hydrogels provide hydrated matrices for regulating protein release near physiological temperature. Building on a previously developed p(ASF-AGE-NIPAAm) hydrogel platform, this study evaluated five formulations with ASF-AGE feed contents of 10.0-57.1 wt% as protein delivery carriers. The hydrogels exhibited lower critical solution temperatures (LCSTs) of 31.8-33.0 °C and reversible swelling-deswelling transitions during cyclic temperature switching between 20 and 40 °C. Increasing the ASF-AGE content decreased the equilibrium swelling ratio from 17.3 to 10.6 g/g, while the bovine serum albumin (BSA) encapsulation efficiency varied between 73.5% and 76.8% and the drug loading capacity decreased from 452.6 to 380.2 mg/g. BSA release was evaluated in phosphate-buffered saline (PBS, pH 7.4) and depended on both temperature and formulation: the apparent cumulative release at the end of the observation period ranged from 37.6% to 54.1% at 20 °C and from 38.4% to 61.2% at 37 °C. Release was higher at 37 °C, coinciding with thermally induced network contraction, and ASF-rich formulations generally released less BSA. These Results define a composition-property-release relationship for p(ASF-AGE-NIPAAm) hydrogels and show that the ASF content can be adjusted to balance protein loading against temperature-responsive release.
