Biomimetic mineralization in layered ZIF-8 enables lipase immobilization for the synthesis of isosorbide dihexanoate.

Kong Lingwen L, Long Huiyi H, Zhou Yuyang Y, Zhou Hua H

Immobilization of lipases in metal-organic frameworks offers a potential route for developing robust biocatalysts for ester synthesis, but achieving efficient enzyme incorporation within structurally favorable hosts remains challenging. Here, we report a biomimetic mineralization strategy based on layered ZIF-8 for immobilization of Candida antarctica lipase B (CALB) and evaluate the resulting biocatalyst for the synthesis of isosorbide dihexanoate (SDH). Under optimized immobilization conditions, namely a CALB dosage of 1 mL, a Zn2+/2-methylimidazole molar ratio of 1:3, and a mineralization time of 6 h, CALB@mZIF-8 exhibited a protein loading of 201.50 mg/g and an activity of 36.71 U/g. The protein loading was higher than that obtained by conventional adsorption-based immobilization using ZIF-8. Applied to SDH synthesis in a 10 mL reaction system containing n-hexane (10%, v/v) as co-solvent, 0.4 g CALB@mZIF-8, an isosorbide-to-hexanoic acid molar ratio of 1:6, agitation at 150 rpm and 40 °C, the SDH yield reached 65.03%. Compared with free CALB, the immobilized biocatalyst exhibited enhanced operational stability and improved tolerance toward different reaction environments. These results demonstrate that layered ZIF-8 generated through biomimetic mineralization provides a tunable platform for lipase immobilization and represents a promising strategy for the synthesis of sterically hindered bio-based esters.

Tunable Acidity of Sulfated Zr-Sn Mixed Oxides for Solvent-Free Sorbitol Dehydration to Isosorbide.

Trivedi Jay B JB, Rana Paresh H PH

One of the most important processes in creating sustainable monomers for high-performance polymers is the catalytic dehydration of sorbitol obtained from biomass to isosorbide. This study investigated a series of sulfated ZrxSn1 - xO2 (x = 0.1-0.9) mixed oxides prepared by coprecipitation method for sorbitol dehydration under solvent-free conditions. Comprehensive evaluation of different characterization techniques, i.e., X-ray diffraction, Fourier transform infrared, transmission electron microscope, N2 physisorption, and NH3-TPD demonstrated that surface acidity, textural characteristics, and catalytic activity were greatly affected by catalyst composition. Catalyst with intermediate Zr content (0.4-0.6) had an ideal balance of weak, medium, and strong Brønsted acid sites, which resulted in complete conversion of sorbitol and higher isosorbide selectivity. 100% sorbitol conversion and 87% isosorbide selectivity were achieved with sulfated (S)-Zr0.5Sn0.5O2 at 220 °C within 3 h, exceeding many reported heterogeneous solid acids catalyst. A minor loss in catalyst activity was observed after four cycles, showing its potential for long-term isosorbide production.

Vascular dementia and vascular cognitive impairment-current treatments and future prospects.

Markus Hugh S HS

Vascular cognitive impairment (VCI) and vascular dementia (VaD) represent a substantial and growing global health burden, yet remain under-recognized and comparatively under-researched. Two broad categories are post-stroke cognitive impairment, and VCI secondary to chronic cerebrovascular disease of which the most common cause is cerebral small vessel disease (SVD). In addition, cerebrovascular pathology frequently coexists with Alzheimer's disease (AD), increasing the probability that AD pathology will manifest as clinical dementia. Current management focuses primarily on prevention through modification of cardiovascular risk factors and lifestyle interventions. Intensive blood-pressure control has shown the strongest evidence, reducing future all-cause dementia while slowing progression of white-matter abnormalities. Multidomain lifestyle interventions incorporating exercise, diet, cognitive training, and vascular risk monitoring may also preserve cognition, although results across trials remain variable. In established VCI and VaD, treatment options are limited; cholinesterase inhibitors and memantine lack convincing evidence of benefit, and no pharmacological therapy has definitively demonstrated symptomatic improvement or delayed disease progression. Advances in understanding SVD and VCI pathophysiology are, however, identifying promising therapeutic targets. Emerging evidence highlights endothelial dysfunction, impaired cerebral autoregulation, increased blood-brain barrier permeability, inflammation, and glymphatic dysfunction as possible mechanisms underlying SVD. Current trials are evaluating treatment approaches based on this framework including cerebral vasodilators such as cilostazol, isosorbide mononitrate and phosphodiesterase-5 inhibitors, anti-inflammatory therapies, glucagon-like peptide-1 receptor agonists, remote ischemic conditioning, and strategies targeting glymphatic function. Improved mechanistic understanding, appropriately targeted interventions, and adequately powered long-term clinical trials are required to translate these emerging approaches into effective treatments for VCI and VaD.

A bio-based superhydrophobic pit and fissure sealant for enhanced stability and anticaries application.

Chen Xu X, Zhang Xiaolei X, Wang Xin X, Zhang Chen C et al.

Dental caries is one of the most prevalent chronic diseases worldwide, with most lesions occurring in occlusal pits and fissures. To prevent occlusal caries, pit and fissure sealants are considered one of the most effective measures. However, their anticaries effect are often compromised by secondary caries resulting from biofilm accumulation. Furthermore, traditional sealants based on bisphenol A glycidyl methacrylate (Bis-GMA) may release bisphenol A (BPA), raising concerns about potential endocrine disruption. In this study, we developed a bio-based superhydrophobic pit and fissure sealant by utilizing isosorbide dimethacrylate (IM) as a bio-based resin matrix, fluorinated methacrylate (FMA) as a low-surface-energy component, and a combination of micro- and nano-sized SiO2 to construct a micro-nano hierarchical surface architecture, after light curing treatment. The bio-based micro-nano structured superhydrophobic pit and fissure sealant (m-n SUP) retained superhydrophobicity after immersion in artificial saliva for 5 days, thermocycling for 5000 cycles, pH cycling for 5000 cycles, tape peeling for 100 cycles, and sandpaper abrasion for 40 cycles. m-n SUP also exhibited favorable mechanical properties and enamel bonding comparable to those of the commercial sealant, together with favorable marginal sealing performance. Moreover, m-n SUP exhibited good cytocompatibility, and markedly reduced salivary mucin adsorption and provided sustained inhibition of the initial adhesion and biofilm formation for five representative oral cariogenic bacteria over 72 h. In a rat model, m-n SUP effectively reduced the incidence and severity of occlusal carie. Overall, this bio-based superhydrophobic pit and fissure sealant can provide a promising bio-based material strategy for pit and fissure caries prevention.

Advancing Epidermal Barrier Resilience in Atopic Dermatitis with Isosorbide Di-Fatty Acid Esters: From Disruption to Restoration.

Chaudhuri Ratan K RK, Sivamani Raja K RK

Atopic dermatitis (AD) is a chronic, relapsing inflammatory skin disease characterized by epidermal barrier dysfunction, immune dysregulation, microbial imbalance, and severe pruritus. Emerging evidence establishes that barrier disruption is a central pathogenic driver capable of initiating inflammatory signaling, neuroimmune activation, and chronic disease instability. This understanding has shifted therapeutic paradigms toward barrier-directed strategies aimed at restoring epidermal resilience. This narrative review evaluates the mechanistic and clinical evidence surrounding isosorbide fatty acid diester molecules-specifically isosorbide dicaprylate (IDC) and isosorbide di-(linoleate/oleate) (IDL)-as a barrier-first approach for AD management. Early in vitro and ex vivo investigations demonstrated that IDC significantly improves epidermal hydration, transepidermal water loss, and the expression of barrier-associated genes linked to epidermal integrity. Subsequent studies showed that IDL expands these effects through coordinated regulation of keratinocyte differentiation, lipid homeostasis, and inflammatory stress pathways. Furthermore, recent mechanistic data highlight synergistic anti-inflammatory and pruritus-modulating effects involving TRPA1-, TRPV3-, and TSLP-associated pathways, while preserving tissue integrity under cytokine-induced stress. Clinically, these findings are supported by randomized studies in pediatric and adult cohorts demonstrating significant reductions in pruritus, favorable Eczema Area and Severity Index (EASI) responses, decreased topical corticosteroid dependence, and a reduction in the relative abundance of Staphylococcus aureus. Collectively, these findings support a barrier-first therapeutic framework in which restoration of epidermal resilience may beneficially influence multiple interconnected pathways involved in atopic dermatitis.

Robust Polyimide/Graphene Composites as Electrically Functional Materials for Space-Compatible Biomedical Applications.

Campanella Martina M, Blondelli Francesca F, Toto Elisa E, Laurenzi Susanna S et al.

Polyimide-based composites with graphene nanoplatelet (GNP) fillers are investigated as multifunctional materials for potential space-compatible biomedical monitoring. These composites are designed to combine the intrinsic high thermal and chemical stability of polyimides, as well as their well-established radiation resistance reported in the literature, with the electrical functionality of graphene. PI/GNP membranes are fabricated by casting dispersions of graphene nanoplatelets within the polyimide solution, using a green and bio-based solvent (dimethyl isosorbide), followed by thermal treatment in vacuum. The effect of UV-C irradiation is investigated as a sterilization-relevant stress condition, showing that the PI/GNP composites retain their main chemical and surface characteristics after exposure. In addition, irradiation is associated with an improvement in the electrical response, suggesting that the conductive network formed by graphene nanoplatelets remains effective after treatment. Overall, this study highlights the PI/GNP composites as robust and electrically functional candidate materials for dry electrode applications in harsh environments. By combining chemical stability, thermal resistance, a stable surface and electrical conductivity, these membranes emerge as promising candidates for future health monitoring applications on Earth and during long-term missions in space.
