Reoviridae-derived short trimerization domain for stabilizing homo-trimerization of vaccine immunogens and therapeutic proteins.

Wei Min M, Bu Qingfang Q, Xiao Jin J, Yuan Lunzhi L et al.

Trimerization motifs play pivotal roles in structural biology and therapeutic protein engineering. Here, we describe a short trimerization motif (rFd1303) derived from the Reoviridae family reovirus σ1 protein. Compared with the widely used T4-Foldon, the rFd1303 increased recombinant protein yields. The rFd1303-fused immunogens of SARS-CoV-2 spike and influenza hemagglutinin elicited antibody responses comparable to T4-Foldon-fused controls in mice. Using this tag, we engineered a trimeric ACE2-Ig fusion protein (TriACE2-Ig) that remained stable after 30 days at room temperature and neutralized a broad range of ACE2-utilizing sarbecoviruses, including 19 SARS-CoV-2 variants, SARS-CoV, and pangolin coronaviruses GD and GX, with higher potency than monomeric ACE2-Ig. In the hamster model challenged with various SARS-CoV-2 variants, our data demonstrated that intranasal TriACE2-Ig administration markedly reduced viral loads, virus-induced body-weight loss, lung pathology, and decreased within-cage virus transmission. These findings highlight rFd1303 as a versatile trimerization platform for vaccine and therapeutic protein development.

Are intervention effects on fruit and vegetable intake maintained after 48 months of follow-up? A randomised controlled community trial.

de Carvalho Maria Cecília Ramos MCR, Lopes Mariana Souza MS, de Freitas Patrícia Pinheiro PP, Lopes Aline Cristine Souza ACS

To assess whether participants from a transtheoretical model-based intervention maintained increases in fruit and vegetables (FV) intake 48 months after baseline. Randomised controlled community trial. A Brazilian primary health care service. Primary health care service users. The control group (CG) received usual care, and the intervention group (IG) received usual care and a 7-month collective intervention to promote FV intake. We analysed individuals with positive changes 12 months after baseline (baseline intake <400 g/d with any increase at 12 months or baseline intake ≥400 g which remained ≥400 g at 12 months, n 2·232). Outcomes were FV intakes at 36 and 48 months after baseline compared to previous intakes to identify maintenance. We used generalised estimating equations models to assess time and intervention effects in follow-up intake. Participants were mostly adult women. We identified decreases of 54.4 g/d in fruit intake and 38.5 g/d in vegetable intake at 36 months and increases of 6.9 g/d in fruit intake and 23.4 g/d in vegetable intake at 48 months. According to adjusted models, at 36 months, CG and IG participants presented similar decreases in FV intakes, which means initial changes in the IG were not maintained. At 48 months, small increases in intakes were also similar between IG and CG. Intervention participants did not maintain initial changes on FV intake in the long term, and boosters might be necessary to maintain initial increases.

Tunable nanofluidic ion gating via in situ salt precipitation-dissolution.

Ji Sungjoon S, Seo Sangjin S, Kim Taesung T

Nanofluidic ion gating underpins both biological signal transduction and iontronic signal processing. However, a simple and reliable mechanism that functions independently of local electrochemical conditions remains unavailable, hindering interference-free integration of artificial iontronic components with biological counterparts. Here, we present a nanofluidic device that enabled ionic gating through pervaporation-driven precipitation and rehydration-driven dissolution (PP&RD) of salt solutions within a nanochannel. Pervaporation induced local salt precipitation in the nanochannel, forming an electrical blockage that suppressed conductance (OFF state), whereas rehydration dissolved the precipitate and restored the conductance (ON state). The ionic gating device exhibited an ON-OFF conductance ratio exceeding 100, fully reversible operation performance, and robust switching across experimental conditions. By integrating two ionic gates in parallel or in series within a single device, we further implement Boolean OR and AND logic operations. PP&RD-based ionic gating thus provided a simple, versatile, and practical foundation for scalable iontronic circuit architecture.

Molecular Confinement Engineering of Mn(II) Halide Scintillators for High-Resolution X-ray Imaging and Three-Dimensional Computed Tomography.

Sun Yating Y, Wang Ting T, Xiong Feixiang F, Yao Yi Y et al.

Metal halide scintillators hold great promise for next-generation X-ray imaging, yet manganese(II)-based halide scintillators suffer from structural instability and thermally activated nonradiative losses arising from their soft lattice nature. Here, we develop a molecular confinement engineering strategy by integrating zero-dimensional CPT2MnBr4 emitters into a cross-linked ionogel network. This strategy simultaneously suppresses moisture-induced degradation and weakens electron-phonon coupling of the embedded emitters. The resulting Mn-IG scintillator exhibits substantially enhanced environmental durability and thermal stability, a high light yield of 19,777 photons Me V-1, and a spatial resolution of 12.7 lp mm-1. Benefiting from high optical transparency and suppressed photon scattering, the Mn-IG scintillator enables high-fidelity X-ray radiography and three-dimensional computed tomography reconstruction. This work establishes molecular confinement engineering as a general strategy for regulating photophysical processes in metal halide scintillators and advancing durable high-resolution X-ray imaging technologies.

Substantia nigra iron-related alterations, white matter disruption, and functional network dysconnectivity in temporal lobe epilepsy: a multimodal neuroimaging study.

Zhao Xu X, Wu Huiting H, Jian Si S, Huang Shanshan S et al.

Although the substantia nigra (SN) has been implicated in temporal lobe epilepsy (TLE) pathophysiology, clinical studies remain scarce. This study aimed to characterize alterations in SN susceptibility, white matter (WM) microstructure, and functional connectivity (FC), as well as their associations with cognitive functions in TLE patients. This study included 20 right-TLE (RTLE), 21 left-TLE (LTLE), and 23 healthy controls (HCs). Multimodal neuroimaging was used to assess the susceptibility, diffusion tensor imaging metrics, and FC of SN subregions. Group differences were examined by analysis of covariance. Exploratory correlation analyses were performed to evaluate associations between imaging metrics and cognitive performance. RTLE patients showed higher susceptibility (post-hoc P = 0.015 vs. HCs; P = 0.002 vs. LTLE) and radial diffusivity (RD) (post-hoc P = 0.014 vs. HCs; P = 0.001 vs. LTLE) in the contralateral substantia nigra pars reticulata (SNr), alongside lower fractional anisotropy (FA) (post-hoc P = 0.009 vs. HCs; P = 0.006 vs. LTLE) in the contralateral substantia nigra pars compacta (SNc). LTLE patients exhibited decreased FA in the ipsilateral SNr (post-hoc P < 0.001 vs. HCs; P = 0.001 vs. RTLE) and reduced connectivity between the left SNc and the right superior parietal lobule (P = 0.026, cluster-level FDR corrected). Susceptibility and RD positively correlated in the contralateral SNr of RTLE cohorts (r = 0.564, uncorrected P = 0.010). Exploratory analyses suggested associations between SNr susceptibility, FA, and cognitive performance in TLE patients (all uncorrected P < 0.05). Our multimodal findings suggest subregion- and laterality-specific alterations in the SN of patients with TLE, with patterns differing between RTLE and LTLE.

Mitigation Strategies: A Standardized Multimodal Pathway to Reduce the Severity of Clinically Relevant Pancreatic Fistula After Pancreaticoduodenectomy.

Casadei Chiara C, Zucchini Valentina V, Giuliani Tommaso T, Solaini Leonardo L et al.

BackgroundPancreaticoduodenectomy (PD) carries significant morbidity, primarily due to clinically relevant postoperative pancreatic fistula (CR-POPF). This study investigated the clinical impact of a multimodal mitigation strategy in patients undergoing PD, encompassing prehabilitation, standardized surgical techniques, and optimized postoperative management. The secondary aim was to identify factors associated with grade C POPF.MethodsA single-center retrospective analysis was conducted using a prospective database of 520 patients who underwent pancreatic surgery at our hospital between January 2016 and December 2025. One hundred eighty-four patients who underwent PD were selected and divided into two cohorts using 2022 as a cutoff, the year in which our multimodal mitigation strategy was introduced.ResultsIn the after-2022 cohort, a significant reduction in CR-POPF was observed (39% vs 55%, P = .036), with specific decrease in grade C POPF cases (5% vs 23%, P < .001). A statistically significant reduction in PPH was also observed in the after-2022 cohort (3% vs 17%, P < .001). Furthermore, in after-2022 cohort, there were a reduction of reoperation rate (8% vs 25%, P = .001) and LOS (23.0 days vs 14.5 days, P < .001). For the secondary aim, the multivariate analysis identified the after-2022 period and the occurrence of cardiac and pulmonary complications as factors associated with grade C POPF (respectively P < .001, P = .017 and P < .001).ConclusionsThis study highlights the potential of a structured, multi-level mitigation strategy in improving outcomes after pancreatoduodenectomy, in particular, the severity of pancreatic fistula and its consequences.
