A Graphene Quantum-Dot Electrometer Array for Sub-Micrometre Imaging of Interfacial Charge Layers in Cryofixed Cells: A Design Analysis.

Celebi Cem C, Ludwig Stefan S, Siddiki Afif A

We analyse, as a design study, a cryogenic sensor that images the electrostatic stray field of a vitrified biological cell with an array of gate-defined quantum dots in gapped bilayer graphene. No device has been fabricated and no transport data are reported. The sensor is a Coulomb-blockade electrometer: each dot converts the offset charge induced on it by the sample into a shift in its resonance, and the steepness of that resonance-a property of the discrete density of states rather than of the device area-sets the gain, so that sensitivity is not lost as the element is made smaller. For a 50 nm dot at 4.2 K we estimate a charge sensitivity of order 10-2 e/√Hz for a charge 50 nm away, limited by an assumed 1/f offset-charge noise; this is a calculated, not a measured, figure. Because a vitrified sample is electroneutral beyond the Debye length, the sensor does not see net charge but the frozen interfacial dipole layers, whose dipole density equals the permittivity times the potential step across each layer. The measured quantity is therefore a band-limited map of lateral variations in the interfacial potential within about 100 nm of the sensor; a laterally uniform layer is invisible. In this map the membrane potential enters with a weight β = Cmt/εwε0 ≈ 0.07 relative to the surface potential of the glycocalyx, so the sensor is primarily a surface-potential imager and only secondarily, through a calibration we specify, a membrane-potential probe. A 10 mV surface-potential contrast at a period of 250 nm exceeds the 1 ms detection limit by a factor of about 4-13, and the 8 × 8 array at 250 nm pitch resolves periods of 250-300 nm. We give the device geometry, the plunge-freezing protocol, the controls and the inversion procedure, and list what remains to be measured.

A Portable UV-LED Dual-Nafion Microfluidic Platform for Label-Free Trace Protein Preconcentration and Absorbance Detection.

Shih Yi-Jia YJ, Chen Yu-Sheng YS, Jen Chun-Ping CP

Sensitive label-free detection of low-abundance proteins requires both effective on-chip preconcentration and a compact, low-noise optical readout for use outside conventional laboratories. Here, we present a portable UV-LED dual-Nafion microfluidic platform that integrates a 280 nm UV-LED, a chromium-coated slot collimator, a dual-Nafion ion concentration polarization (ICP) preconcentrator, and a miniature spectrometer within a 3D-printed enclosure. The symmetric dual-Nafion geometry on a UV-transparent quartz substrate confines the ICP enrichment zone to a fixed 120 × 500 µm interrogation window, enabling spatially stable, label-free on-chip absorbance monitoring at 280 nm. Using bovine serum albumin (BSA) as a model protein, the platform showed a linear absorbance response over 0-60 µM (R2 = 0.9933) and a static-loading limit of detection of 0.91 µM. Under a 30 V bias in 0.3 mM phosphate-buffered saline, representative 60 min experiments produced apparent enrichment factors of approximately 41, 4.5 × 103, and 1.6 × 104 at initial BSA concentrations of 1 µM, 10 nM, and 1 nM, respectively. At 10 pM, a reproducible, voltage-dependent absorbance increase was detected with three independent chips, whereas the 0 V and BSA-free controls remained below the detection threshold. The platform provides a practical approach for label-free trace-protein analysis and a potential upstream module for biomarker-specific assays.

From Choledocholithiasis to Metastatic Pancreatic Adenocarcinoma: A Diagnostic Odyssey Highlighting Missed Red Flags.

Farooq Abdullah A, Saud Jazeela J, Sisay Henok E HE, Hanif Husnain H et al.

Pancreatic adenocarcinoma is one of the deadliest types of cancer, with a very gloomy prognosis, especially if the cancer is at an advanced stage at the time of diagnosis. Among young adults, the vague symptoms that are usual with pancreatic cancer can be confused with those of a benign biliary condition, so the diagnosis is often late. It may be quite challenging to recognize that a bile duct obstruction with jaundice is due to a periampullary tumor, which is similar to a bile duct stone, unless one suspects it very strongly. We present the case of a 38-year-old man who suffered from increasing abdominal pain, significant weight loss, and obstructive jaundice. An initial triphasic CT showed the presence of dilated intrahepatic biliary ducts and a dilated common bile duct (CBD) with duodenal spots that were thought to be passed gallstones. At the same time, a liver lesion was identified as a probable hemangioma. The diagnosis was postponed even though the liver function tests were progressively deteriorating (total bilirubin increased to 10.4 mg/dL and alkaline phosphatase to 1394 U/L). Further endoscopic retrograde cholangiopancreatography (ERCP) revealed a distal CBD stricture without stones; therefore, a plastic stent was placed. No tissue sampling was obtained during ERCP. Five months later, an extremely high carbohydrate antigen 19-9 (CA 19-9) (>500 U/mL) and a repeat triphasic CT showed a 43 × 45 mm pancreatic head tumor with liver and lung metastases. These findings supported a clinical-radiological diagnosis of metastatic pancreatic malignancy, presumed pancreatic adenocarcinoma; histopathological confirmation of the pancreatic or hepatic lesions was not obtained. Subsequent ascitic fluid cytology showed malignant cells consistent with adenocarcinoma, but immunocytochemistry was not available, so the pancreatic primary was not independently established. This case highlights that if a biliary stricture occurs without a stone that can be identified in a patient with progressive obstructive jaundice, the stricture should be considered malignant until demonstrated otherwise. Early obtaining of tissue for diagnosis and appropriate use of tumor markers as supportive rather than definitive tests are the main elements to steer clear of a long diagnostic odyssey.

Transient suction marking for re-identification of a small gastric subepithelial lesion during miniprobe endoscopic ultrasonography.

Soga Koichi K

A 76-year-old woman underwent esophagogastroduodenoscopy, which revealed a 5-mm smooth subepithelial elevation on the anterior wall near the lesser curvature of the upper gastric body with intact overlying mucosa. This elevation became more conspicuous with insufflation. Before deflation, we created a suction mark 3 cm proximal to the lesion. Endoscopic suction was applied for 15 seconds and augmented through the working channel using a 20-mL syringe with the plunger withdrawn by 10 mL. This produced a discrete erythematous mark. Then, the stomach was deflated and filled with 250 mL of water. A 20-MHz miniprobe was advanced using the mark as a mucosal landmark without repeated endoscopic searching for the elevation. Probe insertion began 50 seconds after water instillation, and the first evaluable EUS image was obtained 14 seconds later (total time, 64 seconds). EUS demonstrated an 8-mm lesion arising from the fourth sonographic layer with an internal hyperechoic focus accompanied by acoustic shadowing. The lesion had a smooth outer margin without a cystic component. These findings suggested a small mesenchymal tumor originating from the muscularis propria. A leiomyoma and gastrointestinal stromal tumor were considered in the differential diagnosis. Surveillance endoscopy was planned at 12 months. We applied this principle to re-identify a diminutive gastric SEL after deflation and water filling for miniprobe EUS. Although a single case cannot establish superiority over conventional localization, transient suction marking may provide a simple landmark that does not require a dedicated marking device and may help preserve spatial orientation during EUS assessments.

Iron-Nitride Isomerization as a Deactivation Pathway in Ammonia Oxidation: A Computational Study of Sequential HAT Mechanisms.

Sheng Luo L, Oláh Julianna J

Ammonia oxidation remains a kinetic bottleneck in utilizing ammonia as a carbon-free energy carrier. Iron-based complexes offer a cost-effective alternative to noble-metal catalysts, but use is hampered by low turnover numbers. Here, density functional theory is used to investigate the catalytic activity and deactivation pathways of Fe2+/Fe3+ complexes bearing cis-ammonia ligands. We confirm the ground-state electronic structures of these complexes, then establish an accurate SMD/DRACO protocol─using a van der Waals cavity with Gaussian-smeared surface charge discretization─that predicts redox potentials with mean error below 0.013 V versus experiment. Thermodynamic evaluation of sequential hydrogen atom transfer (HAT) steps reveals a "peaked" bond dissociation free energy profile. Crucially, the exergonic third HAT step yields not the anticipated nitrido species, but a novel isomer in which the nitride inserts into the Fe-C bond. This Fe-N-C isomer is a deep thermodynamic sink, trapping the catalyst in an inactive state. A similarly low-energy Fe-N-C isomer exists for the imido intermediate but is kinetically inaccessible. These results indicate that Fe-C-containing complexes are unlikely to serve as efficient AO catalysts, as a high-valent iron center with a multiply bonded nitrogen ligand renders the Fe-C bond prone to alkyl migration, a facile deactivation pathway.

Ferrocene-Branched Polymers via a Ugi Multicomponent Reaction for an Integrated and Reagentless Amperometric Glucose Biosensor.

Urraca Jorge J, Zurita Isabel I, Krznaric Marija M, Martínez Tatiana T et al.

A Ugi multicomponent reaction was employed to synthesize a ferrocene (Fc)-branched polyethyleneimine derivative. This polymer was evaluated as a redox-active matrix for the fabrication of an electroanalytical disposable glucose biosensor based on a carbon PE-printed electrode functionalized with glucose oxidase as a biorecognition element. The resulting biosensor displayed a linear response up to 35 mM glucose at pH 6.5, with a limit of detection of 33 µM. The enzyme biosensor reached 95% of the steady-state current within approximately 50 s and showed a sensitivity of 0.92 µA/mM. In addition, this analytical device exhibited high selectivity, good reproducibility, and excellent storage stability, retaining strong analytical performance after 45 days at 4 °C. The proposed electrochemical biosensor was successfully applied to glucose determination in red and white wine samples.
