Purification of Bioactive Natural Antibodies to PD-1 Known as Regulatory Rheumatoid Factor from Human Blood Plasma.

Menshikova Daria D, Terentiev Alexey A, Ivanov Pavel P, Cherednichenko Alexandr A et al.

Regulatory rheumatoid factor (regRF) is a recently discovered population of natural anti-idiotypic antibodies carrying a shared paratope specific to the PD-1 molecule. Normally, regRF exerts a cytotoxic effect on PD-1-expressing T lymphocytes, including autoreactive ones, thereby controlling their expansion. The specificity for PD-1 indentifies regRF as a promising therapeutic agent for suppressing autoimmune reactions via the PD-1 pathway. The aim of this study was to develop gentle methods for regRF isolation from healthy blood plasma, which would preserve the bioactivity of regRF. Ammonium sulfate precipitation was used for crude regRF purification. A search for suitable affinity sorbents was carried out. The purified regRF was tested for its bioactivity. Ammonium sulfate precipitation at 35% saturation, followed by extraction with 28% saturated ammonium sulfate solution, allowed the separation of regRF from a portion of non-target IgG and high-molecular-weight fibrinogen. RegRF showed high affinity for Cibacron Blue and no binding to Protein A. Sequential affinity chromatography on immobilized Cibacron Blue and Protein A enabled the purification of regRF from non-target immunoglobulins and low-molecular-weight fibrinogen. The purified regRF samples showed binding to PD-1 and reduced the PD-1+CD4+ T lymphocyte count in vitro. Thus, we have identified sorbents and conditions for the purification of biologically active regRF.

Xanthan Gum/Lignin Double-Interpenetrating Network Cryogels: Preparation, Characterization, and Water Purification Performance.

Lazar Maria Marinela MM, Humelnicu Doina D, Raschip Irina Elena IE, Avram Raluca-Elena RE et al.

The development of multifunctional, bio-based materials capable of simultaneously addressing microbial contamination and inorganic pollutants is of increasing interest for sustainable water purification. In this study, xanthan gum (XG)/lignin(LN) double-interpenetrating network (DN) cryogels were prepared and investigated as porous, multifunctional materials for antimicrobial applications and water purification. The cryogel architecture combines the abundant oxygen-containing functional groups of XG and LN with an interconnected porous network generated under cryogenic conditions. The prepared networks were comprehensively characterized using Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), dynamic vapor sorption (DVS), swelling analysis, and uniaxial mechanical testing to evaluate their structural, morphological, physicochemical, and mechanical properties. Their Cu2+ sorption performance was subsequently assessed by atomic absorption spectroscopy (AAS), and the experimental equilibrium data were analyzed using the Langmuir, Freundlich, and Sips isotherm models. The single-network (SN) XG-based materials exhibited Cu2+ sorption capacities of 62.13-109.81 mg/g, whereas the DN cryogels showed higher capacities of 111.87-120.84 mg/g. The Sips model provided the best description of the equilibrium data, with (R2) values of 0.991-0.994 and substantially lower (χ2) values than the Langmuir and Freundlich models, indicating heterogeneous sorption sites with finite saturation capacity. Importantly, the Cu2+-loaded SN and DNs exhibited sorption efficiency toward sulfate ions, demonstrating their potential for sequential contaminant removal. Sulfate sorption capacities reached 694.53 mg/g for the SN.X2/Cu2+ composite and up to 812.53 mg/g for the DN.XL3/Cu2+ composite. In addition, the prepared materials demonstrated antimicrobial activity against two Gram-negative (Escherichia coli, Salmonella Typhimurium) and two Gram-positive (Staphylococcus aureus, Listeria monocytogenes) bacterial strains. The results demonstrate that XG/LN DN cryogels combine effective metal-ion and sulfate sorption with antimicrobial activity, providing promising bio-based platforms for water purification and potentially extending the functional lifetime of the material through sequential contaminant removal.

Insights into the Protonation State and Counterion Interactions of Linear Polyethyleneimine Materials.

Byrne Agustín J AJ, Vladimir Daniel D, Garro-Linck Yamila Y, Copello Guillermo J GJ et al.

Polyethyleneimine (PEI) polymers are widely used in the synthesis of nanoparticles, hydrogels and composite materials for biotechnological and environmental applications. However, the solid-state structural organization of linear PEI (L-PEI) materials, and how it is influenced by molecular weight, protonation state and counterion type, have not yet been comprehensively studied. In this work, solid-state nuclear magnetic resonance (ss-NMR), X-ray powder diffraction (XRD), small-angle X-ray scattering (SAXS) and time-domain NMR (TD-NMR) experiments were combined with theoretical calculations to analyze the local chemical environments, crystalline and nanoscale organization, and molecular mobility of L-PEI materials with different molecular weights. The analyzed materials were the free base (PEI·FB), the hydrochloride (PEI·HCl), the trifluoroacetate (PEI·TFA), and the sulfate-containing forms of L-PEI (PEI·Na2SO4 and PEI·H2SO4), as well as an intermediate protonation state obtained by the heterogeneous treatment with HCl-saturated isopropanol. Proton, 13C or 15N ss-NMR studies evidenced different chemical environments associated with changes in protonation state and counterion identity. Furthermore, XRD and SAXS showed that these variables, together with molecular weight and sample preparation conditions, influenced the crystalline arrangement and characteristic nanoscale organization of the polymer chains. Moreover, TD-NMR revealed the presence of three dynamically distinct proton populations, assigned to rigid, interfacial, and mobile domains, whose relative contributions and relaxation behavior varied among the PEI materials. Finally, high-resolution magic-angle spinning (HRMAS) NMR experiments were used to study different PEI-gel materials and films as well as the crosslinking of protonated PEI and sulfate ions.

Tailoring phase modifiers for selective uranium extraction: conventional and phosphate ores.

Gautier M M, Lin J J, Guerinoni E E, Daronnat L L et al.

Selective uranium extraction from sulfate and phosphate-based leachates remains challenging due to co-extraction of impurities such as Mo, Zr, and Fe, which limits process efficiency and economic viability. This study investigates how the nature and concentration of phase modifiers influence uranium selectivity across three mining systems: two conventional uranium ores and one phosphate-associated ore. U, Mo and Zr were quantified by ICP-OES, while Fe was analyzed by gamma-ray spectrometry. Results show that phase modifier effects on selectivity are highly system dependent. In AMEX-based sulfate systems, increasing EXXAL 10 enhances selectivity through systematic improvements in extraction performance: in Mine A, U/Zr increases from 50 to 221 at 50 vol% (vs. 5 vol% industrial conditions), while in Mine B, U/Mo increases from 377 to 1459 at 22 vol%, with optimal U/Zr selectivity between 10-50 vol%. In contrast, the phosphate-based Mine C exhibits a weakly structured organic phase, with dimer-dominated extractant species and limited sensitivity to phase modifier addition; optimal selectivity is achieved at low modifier content, where 2 vol% EXXAL 13 increases the U/Fe ratio from 61 to 214. These results highlight the importance of system-specific phase modifier optimization for maximizing uranium selectivity. SAXS analysis further reveals that extractant molecules self-assemble into reverse aggregates whose structure and inter-aggregate interactions are strongly influenced by phase modifier concentration. Crucially, these structural modifications directly correlate with selectivity variations, demonstrating that phase modifier effects are fundamentally mediated through changes in extractant aggregation in the organic phase.

Crystal Morphology of Luminescent Microcrystals Based on Eu(III) Coordination Polymers Changed by Surfactant Solutions.

Enokido Masaki M, Wang Mengfei M, Kitagawa Yuichi Y, Hasegawa Yasuchika Y

The morphological control of Eu(III) coordination polymers with intermolecular interactions between polymer chains was demonstrated with surfactants as molecular modulators. [Eu(hfa)3(m-dpb)]n coordination polymers (hfa; hexafluoroacetylacetonate, m-dpb; 1,3-bis(diphenylphosphoryl)benzene) showed crystal growth in CTAB/SDS solution (CTAB, cetyltrimethylammonium bromide; SDS, sodium dodecyl sulfate). The crystal morphology of [Eu(hfa)3(m-dpb)]n was changed under the synthetic conditions with various types of cationic surfactants (N+(CH3)3CnH2n+1; n = 8, 12, and 16). The Eu(III) coordination polymer crystals were observed as plates and slender rod shapes. Their crystal morphologies suggest that the crystal growth along the chain growth direction is faster than that along the chain stacking direction.

Emergent challenges: Eclampsia case series in primigravida Rohingya refugees of Malaysia.

Zakour Khadari Mohamad M, Zainal Hadzliana H, Athirah Daud Nur Aizati NA, Sha'aban Abubakar A et al.

Eclampsia, a serious and life-threatening complication of pregnancy, affects women in low-income and vulnerable populations. This case series describes the clinical management of two young Rohingya refugee women, aged 17 and 21, who presented with eclampsia at Seberang Jaya Hospital, Penang, Malaysia. Both patients reported prodromal symptoms of severe headache and visual disturbances before experiencing generalized tonic-clonic seizures. Treatment involved intravenous magnesium sulfate for seizure control and labetalol for blood pressure control, in accordance with local clinical guidelines. Notably, the 17-year-old patient's recovery was complicated by concurrent dengue fever, which presented additional management challenges. Neonatal outcomes varied significantly: the first infant had an Apgar score of 3 at 1 min and a birth weight of 1.7 kg, while the second infant had a healthier outcome, with an Apgar score of 9 and a birth weight of 3 kg. These cases emphasize the critical need for timely diagnosis, standardized treatment protocols, and comprehensive maternal and neonatal care in managing eclampsia among vulnerable populations.
