Variability of natural killer activity during immunotherapy.

In patients with multiple sclerosis, chronic active hepatitis B, and relapsing genital herpes integral parameters of variability in natural killer activity were higher than in donors. In the dynamics of immunotherapy with IFN preparations (chronic active hepatitis B) and IFN inductor ridostine (relapsing genital herpes) these parameters decreased and approached the normal values. The possibility of using variability of natural killer activity for evaluation of cell function and prediction of the efficiency of immunotherapy is discussed.

[An intensification of antiviral and interferon effects of ridostin (an interferon inducer) by cationic liposomes in vitro and in intranasal administration].

Combined application of ridostine with catonic liposomes was shown to essentially enhance the interferon-inducing and antiviral activity of the former in experiments with cell cultures L-929, which is apparently related with an improved efficiency of intracellular delivery of dsRNA. A comparative study demonstrated that ridostine, when combined with liposomes, is needed by 10(3)-10(4) times less as when it is used alone. A pretreatment of the cellular monolayer by cationic liposomes contributes also to enhancing the activity of ridostine, which can be explained by an enhanced permeability of cells for dsRNA holding on-for as long as 30 minutes after the removal of liposomes from the liquid culture. A separate successive administration of, first, liposomes and, then, of ridostine in BALB/c mice (20 mg/kg) leads to a more intensified induction of interferon in the upper respiratory tract tissues as compared with the administration of ridostine alone.

[Effectiveness of the interferon inducers ridostin and camedon in prevention and treatment of experimental alpha- and flavivirus infections].

Antiviral activity of rybamidil (virasol) and interferon inducers ridostine and camedone developed in Russia and introduced in practical medicine has been studied in mice with experimentally induced arboviral infections, flavivirus (tick-borne encephalitis-TBE, and yellow fever-YF) and alphavirus ones (Western and Eastern equine encephalomyelitis-WEE and EEE). Rybamidil injected subcutaneously proved to be ineffective both for prevention and treatment of these infections. Early interferon inducers ridostine and camedone (larifan) with a peak of interferon production 4 hours after injection, when used in doses of 5 and 150 mg/ kg, respectively, had a clear-cut prophylactic antiviral effect if injected 24 or 4 hours before infection. Ridostine had a distinct antiviral effect, providing up to 40% protection from TBE, YF, and WEE.

[The experimental efficacy of vaccination with a recombinant yeast vaccine against hepatitis B in combination with immunomodulators].

Experiments in BALB/c mice were carried out to study a number of immunomodulators in combination with vaccination using a recombinant yeast hepatitis B vaccine. The use of poludan, ridostine, amixine, larifan, myekonide significantly enhanced specific antibody production and at least doubled cell-mediated immune response by 14 and 28 days postvaccination. The above-mentioned immunomodulators are planned to be used in vaccination with the recombinant yeast vaccine against hepatitis B.

[Effectiveness of Soviet derivatives of phosphonic acid and their combination with interferon inducers in cell cultures and in a model of herpetic meningoencephalitis in mice].

Nationally synthesized chemopreparations: phosphonoacetic (PAA), phosphonoformic (PFA) acids and acycloguanosine (Acg) exhibit a marked antiherpetic effect in cell cultures and marked protective effect in herpes meningoencephalitis in mice induced by intraperitoneal inoculation of herpes simplex virus type 1 (HSV-1) (43% PFA, 33% PAA, 25% Acg). Both in vitro and in vivo (mouse meningoencephalitis), PFA (its trisodium salt) on the whole proved to be less toxic than PAA but exerted a higher or similar antiherpetic effect. The combined use of pyrophosphate analogues (PAA, PFA) with Acg is more effective that their use separately both in vitro and in herpes meningoencephalitis in mice an produces an additive effect or one similar to it. Systemic inoculation of interferon inducers, lafarine or ridostine, is effective in herpes meningoencephalitis in mice induced by intraperitoneal inoculation of HSV-1 (the protective effect 33% and 26%, respectively). The combined use of ridostine and PAA in herpes meningoencephalitis in mice led to synergistic effect.
