A Three-Dimensional Biomimetic In Vitro Model to Simulate Schwann Cell-Mediated Peripheral Nerve Repair.

Peripheral nerve injuries represent significant clinical challenges, often resulting in lifelong motor function loss and disability. Evidence suggests regenerating axons cannot cross nerve gaps without Schwann Cell (SC) assistance. However, FDA-approved biomaterial conduits for peripheral nerve repair lack bioactivity and structural complexity needed to facilitate SC migration. To address this, we developed a three-dimensional biomimetic in vitro model to simulate complex cellular interactions within the nerve bridge. The model features lyophilized hydrogel bioscaffolds with longitudinal channels to recapitulate the nerve microenvironment. To encourage directional SC dispersion, the top ~30% of the bioscaffold was conjugated with macrophage inflammatory protein-1α (MIP-1α). Rat SCs were seeded within no-MIP-1α- and MIP-1α-conjugated bioscaffold channels as spheroids and cultured for nine days. Histology demonstrated MIP-1α conjugation retained more SC spheroids during culture with greater cellular distributions. SC spheroid culture in MIP-1α-conjugated bioscaffold resulted in enhanced paracrine signaling characterized by increases in VEGF, ICAM-1, IL-6, and CINC-1 production, alongside downregulation of IL-1β, IL-10, and IL-13. The SC-derived pro-inflammatory and pro-angiogenic mediators did not inhibit NSC-34 motor neurite extension compared to controls. This study establishes an in vitro model that serves as both a screening platform and mechanistic tool, advancing our understanding of peripheral nerve repair.

Dextrin Palmitate and Disteardimonium Hectorite Construct a Gel-like EHMC Matrix: Enhanced UVB Photoprotection and Plasma Exposure Modulation.

2-Ethylhexyl-4-methoxycinnamate (EHMC) is among the most widely adopted organic UVB filters in commercial sunscreens. Nevertheless, its practical application potential is limited by unfavorable formulation compatibility and safety risks stemming from systemic exposure after topical administration. In this study, an oil-continuous structured gel matrix consisting of EHMC, disteardimonium hectorite (DDH) and dextrin palmitate (DP) was constructed to enhance UVB photoprotection and modulate the plasma exposure profile of EHMC following topical application. Comprehensive characterizations including rheology, XRD, Raman spectroscopy, FTIR spectroscopy, TGA and SEM collectively revealed that the combined incorporation of DDH and DP facilitates matrix structural rearrangement, enables EHMC to bind within the structured network, and promotes the formation of more intact continuous surface films. In vitro SPF assays demonstrated that the finished topical formulation SC-4 delivered superior UVB blocking efficacy compared with the EHMC-only control SC-1; furthermore, SC-4 exhibited improved short-term physical stability under the preset thermal and centrifugal acceleration test conditions. Follow-up skin safety assessments, mass spectrometry imaging (MSI) and pharmacokinetic assays verified that SC-4 elicited no remarkable acute skin irritation across all experimental conditions. Relative to SC-1, the reference formulation with EHMC as the sole UV filter, SC-4 displayed weaker EHMC-related distribution signals in skin tissues, accompanied by lower early plasma EHMC concentrations and a slightly lower AUC0-48h trend. Collectively, these findings indicate that DDH/DP co-assembly serves as a viable matrix-structuring strategy to modulate EHMC-related skin distribution and early plasma exposure. Further research into UVA blocking performance, photostability, skin retention and transdermal permeation profiles, as well as long-term storage stability, is required to advance the development of broad-spectrum sunscreen formulations built on this novel matrix platform.

Scratching the Surface: Lipidomic Profiling of the Stratum Corneum in the Search for Pruritogens in Cholestatic Liver Diseases.

Pruritus is a debilitating symptom frequently affecting patients with cholestatic liver disease, often resistant to conventional antipruritic therapies. The pathogenesis of cholestatic pruritus (CP) is multifactorial, implicating not only bile acids but also a complex array of other potential pruritogenic mediators and neural signaling pathways. Identification of the exact pruritogen has been elusive, and gaps remain in understanding the pathogenesis of pruritus, so developing targeted treatments is critical. The stratum corneum (SC), the outermost lipid-rich layer of the skin, may act as a reservoir for circulating pruritogens, offering a novel window to explore the pathogenesis of CP. Recent advancements in lipidomics and non-invasive tape stripping have enabled detailed profiling of SC lipid alterations in disease states. In this review, we synthesize the current understanding of CP and its candidate pruritogens and describe state-of-the-art approaches for SC lipid analysis, combining tape stripping to sample the skin surface with mass spectrometry-based lipidomics. Finally, we summarize cutaneous molecular findings and discuss how these techniques are facilitating biomarker discovery and informing therapeutic development. This review discusses the paradigm shift from a single-molecule perspective to a more integrated view of CP as a product of complex mediator interactions and highlights the potential of SC profiling to uncover novel targets for intervention strategies.

Associations Between Weight Status, Peer Victimization, and Mental Health in Clinically Referred Children: A Cross-Sectional Study.

To examine peer victimization, internalizing symptoms, and psychiatric disorders in underweight and obese clinically referred youth versus normal-weight peers. Cross-sectional study of 161 children aged 7-17 (51 underweight, 57 normal-weight, 53 obese). Victimization was measured using the Olweus Bully/Victim Questionnaire, depressive and anxiety symptoms with the CDI and STAI-C trait, and psychiatric diagnoses with the K-SADS-PL. Peer victimization and anxiety disorders were higher in underweight and obese than normal-weight children (p < .001 and p = .005, respectively). Trait anxiety symptoms were elevated in both groups (p < .001), whereas depressive symptoms did not differ. Victimized youth had higher rates of anxiety (p = .009) and depressive disorders (p = .001). In adjusted models, underweight predicted anxiety disorder (OR 5.03, p = .010), underweight (OR 4.19, p = .003) and obesity (OR 9.00, p < .001) predicted victimization, and victimization predicted depressive disorder (OR 3.03, p = .016). Weight extremes in clinically referred children are linked to peer victimization and internalizing problems, supporting routine psychosocial screening in clinical practice.

Correlation Between Multifocal Electroretinogram and Optical Coherence Tomography Findings With Visual Acuity in Patients With Macular Dystrophy.

To correlate the optical coherence tomography (OCT) findings and multifocal electroretinogram (mf-ERG) responses with visual acuity (VA) in patients with macular dystrophies. A cross-sectional study including 62 eyes of 31 patients diagnosed with macular dystrophy was conducted. All participants underwent assessment of corrected-distance VA, mf-ERG responses (data analysis were restricted to ring 1), and OCT parameters, including central macular thickness (CMT) and the integrity of the inner segment/outer segment (IS/OS) junction. The correlations between VA with mf-ERG responses and OCT parameters were analyzed. A statistically significant moderate correlation was found between corrected-distance VA and CMT (r = 0.497, P < .001), the mf-ERG amplitudes of N1 (r = 0.469, P < .001), N2 (r = 0.517, P < .001), and P1 (r = 0.494, P < .001), the mf-ERG latencies of N2 (r = -0.301, P = .017), and the integrity of the IS/OS junction (r = -480, P < .001). There was a statistically significant weak negative correlation between corrected-distance VA of the studied eyes with mf-ERG latencies of N1 (r = -0.269, P = .034) and P2 (r = -0.283, P = .026). Regression analysis showed that CMT and IS/OS junction integrity were the most significant predictors of VA, alongside mf-ERG amplitudes and latencies. CMT and the integrity of the IS/OS junction are strong structural indicators for visual function in patients with macular dystrophies. Moreover, mf-ERG is considered to play a crucial role in identifying early functional impairment before irreversible anatomic changes occur.

Developmental trajectories of self-efficacy and its predictors among people with severe disabilities.

The aim of this study was to examine the developmental trajectories of self-efficacy in individuals with severe disabilities and to identify predictive factors accounting for variations. This research utilized latent growth curve modeling to analyze data from the first (2016), third (2018), and fifth (2020) waves of the second Panel Survey of Employment for the Disabled, conducted by the Korea Employment Agency for Persons with Disabilities. The analytic sample consisted of 1,285 working-age adults (19-64 years) with severe disabilities, as defined by the Act on Welfare of Persons With Disabilities in South Korea. Overall, self-efficacy among individuals with severe disabilities demonstrated a steady increase over time. A significant negative covariance was observed between the initial level of self-efficacy and its rate of change, indicating that individuals with lower initial self-efficacy experienced more rapid growth over time. With respect to predictors, higher initial self-efficacy was positively associated with family support for employment (β = .192, p < .001), participation in social activities (β = .139, p < .001), and the extent of interpersonal relationship maintenance (β = .598, p < .001). In contrast, the rate of change in self-efficacy was negatively associated with family support for employment (β = -.134, p < .01), the number of job aids (β = -.123, p < .001), and the extent of interpersonal relationship maintenance (β = -.377, p < .001). These patterns indicate more gradual increases among individuals with higher initial levels of self-efficacy and steeper growth trajectories among those with fewer initial resources. The findings suggest that although individuals with severe disabilities who lack initial resources may begin with lower levels of self-efficacy, they can achieve substantial developmental gains through environmental support and accumulated experience. Self-efficacy thus appears to function as a malleable psychological resource within rehabilitation contexts, highlighting the potential of targeted environmental and intervention-based strategies to promote adaptive developmental trajectories.
