Integration of the Triboelectric Effect into a Yolk-Shell Conjugated Microporous Polymer for Enhanced Artificial Photosynthesis.

Conjugated microporous polymers (CMPs) have demonstrated considerable potential in the field of photocatalytic organic synthesis. However, the issue of intrinsic photogenerated carrier recombination severely limits their potential applications. Herein, a strategy for boosting the photocatalytic performance of CMP through the triboelectric effect is proposed. Specifically, the benzothiadiazole-involved CMP (BTTP) with inherent triboelectric character is processed into yolk-shell SiO2@BTTP materials, in which the BTTP and SiO2 function as tribo-negative and tribo-positive materials, respectively. The dual triboelectric effects occurring on both the internal and external surfaces of the shell are capable of inducing the formation of an additional built-in electric field within the BTTP photocatalyst, which effectively promotes the separation of photo-generated carriers, significantly enhancing photocatalytic performance. In the photocatalytic aerobic oxidation of the representative methyl phenyl sulfide, the yolk-shell SiO2@BTTP exhibits an ultrahigh sulfoxide yield of ∼399.6 mmol·g-1·h-1 under optimized conditions, accompanied by exceptional selectivity and cycling stability. Furthermore, this enhancement arising from the triboelectric effect is also observed in the selective photocatalytic oxidation of other organics by BTTP, including sulfide derivatives, benzyl alcohol and benzylamine. The findings of this work on utilizing intrinsic friction characteristics to boost the photocatalytic performance of CMP provide alternative avenues for the development of high-performance CMP-based photocatalysts, which will greatly unlock the application potential of such semiconductors in the field of artificial photosynthesis.

Relative Bioavailability of Once-Daily Clonidine XR Oral Suspension Versus Twice-Daily XR Tablets in Healthy Volunteers.

To assess the bioequivalence of once-daily clonidine extended-release (XR) oral suspension (OS) to twice-daily clonidine XR tablets (TABs), which are approved for attention-deficit/hyperactivity disorder (ADHD). Study 1 (n = 20) evaluated an equivalent daily dose of clonidine XR OS versus TABs and clonidine XR OS under fed versus fasted conditions. Outcomes included the maximum plasma concentration (Cmax), the area under the analyte versus time curve (AUC) from 0 to the last analyte concentration (AUCt) and to infinity (AUCinf), and the half-life (Thalf). Study 2 (n = 19) evaluated AUC, Cmax, and the minimum concentration (Cmin) at steady state (SS). The ratios of geometric means and corresponding 90% CIs of pharmacokinetic parameters for each treatment were compared to the US Food and Drug Administration definition of bioequivalence (80%-125%). In Study 1 (mean [SD] age, 42 [11] years; 30% female; 10% Asian, 25% Black or African American, 5% Multiracial, 60% White; 40% Hispanic or Latino), the ratios (90% CIs) of clonidine XR OS to TABs were within the bioequivalence range accepted by the FDA for Cmax (95.6 [89.8, 101.8]), AUCt (97.2 [91.6, 103.1]) and AUCinf (96.1 [89.4, 103.4]); similar results were observed for clonidine XR OS under fed versus fasting conditions. The median (range) Thalf was 12.8 (8-24) hours. In study 2 (mean [SD] age, 43 [11] years; 42% female; 47% Asian, 26% Black or African American, 0% Multiracial, 26% White; 0% Hispanic or Latino), AUCt,ss and Cmax,ss were both within the acceptable range (97.7 [93.4, 102.1] and 107.9 [103.8, 112.2], respectively). The Cmin,ss for clonidine XR OS was ~26% lower than clonidine XR TABs (74.0 [69.3, 79.0]) but this difference was not considered clinically meaningful by the FDA. These results demonstrate that clonidine XR OS is bioequivalent to clonidine XR TABs and could fulfill a treatment gap as a liquid, once-daily, non-stimulant ADHD treatment option.

Effect of Bacteriophage Administration Route on Phage Localization in a Rat MRSA Implant-Associated Infection Model.

Background/Objectives: Implant-associated infections are challenging conditions in orthopedic surgery. This experimental study aimed to evaluate phage localization within infected tissues following different routes of administration. Methods: An implant-related infection model was created using methicillin-resistant Staphylococcus aureus (MRSA) in twenty-four rats. Subjects were randomly divided into four groups depending on the bacteriophage administration route. Three rats were designated as the control group. Phage suspension was applied intraperitoneally, intravenously, orally and locally at 0.1 mL/day of 1 × 108 PFU/mL suspension for three consecutive days. In the control group, intravenous, intraperitoneal and oral phage suspensions were administered separately at the same dose for 3 days. After completion of the experiment, tibia samples were taken in the experimental group. Additionally, liver, kidney, stomach, brain, heart muscle and striated muscle tissue samples were taken from the three subjects in the control group. Results: In the control group, unconfirmed phage-like structures were incidentally observed in some mitochondria of renal proximal tubular epithelial cells on transmission electron microscopy. In the experimental group, there was a strong positive linear relationship between the total number of bacteria and the number of bacteriophage clusters, independent of the groups. Conclusions: Bacteriophage clusters were detected in infected tibial tissues after all administration routes, suggesting phage localization at the infection site. Unexpected phage-like clusters were observed within mitochondria of proximal tubular epithelial cells in the control animals. This finding should be regarded as an unconfirmed incidental finding requiring further validation.

A high-throughput, plate reader-based method for the assessment of oxidative stress in suspension mammalian cells using CellROX Green.

The measurement of intracellular reactive oxygen species (ROS) levels provides critical insights into cell health and pathophysiology. Here, we present a semi-quantitative, plate reader-based protocol using CellROX Green that can be used for the real-time measurement of general ROS levels of viable suspension mammalian cells. With menadione being the oxidative stress inducer, a cell concentration of 100,000 cells per well, a CellROX Green concentration of 10 µM, and a CellROX Green incubation time of 1 h was shown to provide the most accurate measurement of intracellular ROS levels in viable human monocyte leukemia (THP-1) cells. Unlike plate reader protocols using other general ROS-sensitive fluorogenic probes, this plate reader protocol eliminates the washing step of the unbound probe prior to measuring fluorescence, making it more suitable for suspension cells. Using the presented protocol, the fluorescence of up to 96 samples can be measured in under 1 min, making it ideal for rapid screening applications where moderate to large differences in oxidative stress levels are to be measured.

Numerical Solution of the Problem of Relaxation Filtration of a Suspension Through a Radial Filter at a Constant Flow Velocity.

This paper investigates the relaxation filtration of a suspension through a radial filter surface under conditions of constant flow velocity. A mathematical model for relaxation cake growth is formulated based on the liquid-phase continuity equation, Darcy's relaxation law, and constitutive relations for both compressive and liquid pressures. The resulting governing equation is a nonlinear partial differential equation for the compressive pressure, complemented by a Stefan condition that characterizes the motion of the cake-slurry interface. The moving-boundary problem is solved numerically using a finite difference method employing a coordinate-based front-tracking technique combined with iterative procedures. The numerical results demonstrate the influence of relaxation effects on cake formation. Increasing the relaxation time slows the compaction process, thereby maintaining higher porosity and promoting accelerated growth of the cake layer thickness.
