Nose-to-brain delivery of IRX4204 by cell-adhesion peptide-functionalized gemini surfactant-phospholipid nanoparticles.

Efficient delivery of hydrophobic therapeutics to the central nervous system (CNS) remains challenging due to poor solubility, limited blood-brain barrier permeability and systemic toxicity. Here, we report the development of gemini surfactant-phospholipid nanoparticles (GPNPs) for intranasal delivery of the highly potent and preferential retinoid X receptor agonist IRX4204. Physicochemical characterization demonstrated stable nanoparticles (60 nm, +40 mV) with high encapsulation efficiency and long-term stability. Dynamic light scattering, transmission electron microscopy and small-angle X-ray scattering revealed that optimal drug loading in GPNPs is ≤0.5 mg mL-1. The formulations showed the coexistence of two structurally distinct particle populations, flattened micelles and larger vesicular structures. In vivo studies in mice demonstrated that intranasal administration of IRX4204-GPNPs (0.12 mg kg-1) achieved higher CNS concentrations than a 100-fold higher oral dose (12 mg kg-1), with sustained distribution across various CNS regions and enhanced striatal accumulation. Compared to oral IRX4204 suspension administration, intranasal delivery of IRX4204-GPNPs achieved about 1.4-fold higher CNS drug concentrations, 136-fold greater delivery efficiency (0.19% vs. 0.0014% of dose) and 73.5% direct transport percentage (DTP), accompanied by markedly reduced plasma and liver exposure. Pharmacodynamic evaluation confirmed sustained activation of RXR-responsive genes, including SCD1 and LXRα, following intranasal nanoparticle delivery. These findings demonstrate that optimized GPNPs enable efficient CNS targeting and support their use as a promising intranasal platform for delivery of RXR agonists and other hydrophobic therapeutics.

Velociraptor Machine Learning Quantifies Similarity to Known Cell Types and Matches Cells Across Flow and Imaging Cytometry Platforms.

Suspension flow cytometry enables high-throughput cellular profiling at the single cell level, but these data lack positional information. Conversely, tissue-based imaging cytometry techniques reveal a cell's location within the tissue architecture and can provide insight into cell biology. It would be especially valuable if data analysis tools could incorporate data from imaging and flow cytometry platforms to gain complementary strengths when quantifying features of cells and populations. We hypothesized that per-cell Marker Enrichment Modeling (MEM) might provide a way to register cells between flow and imaging cytometry analysis. Here, we developed the Velociraptor machine learning workflow for cross-platform cytometry analysis. Velociraptor begins with a graph-based implementation of MEM to calculate per-cell quantitative phenotype labels. With this information, Velociraptor can then quickly calculate similarity between each cell's phenotype and search terms describing established cell types, cells of interest, or cells observed in other samples. Velociraptor was effective in registering cells within and between cytometry platforms. Integrated identification of cell populations was tested in several challenges, including comparisons of high dimensional datasets from cancer and immunology. Tested instrument types included imaging mass cytometry (IMC), cyclic immunohistochemistry (cycIHC), suspension mass cytometry (CyTOF), and suspension spectral flow cytometry (SFC). Between IMC and CyTOF, a comparison across imaging and flow cytometry platforms that use the same mass tag probes, Velociraptor accurately identified and registered immune cell types (median F1-measure of 0.81). Between SFC and CyTOF, a comparison of two fundamentally different probe types-fluorophores and metal tags-in suspension flow cytometry, Velociraptor was even more accurate at identifying and registering cells (concordance correlation coefficient of 0.99). Velociraptor was especially useful in heterogeneous samples where individual cells diverged in phenotype from the bulk population. In IMC imaging of human breast cancer, a previously unappreciated tumor cell subset was revealed by Velociraptor, characterized as CD15+, and validated as spatially segregated to the tumor core. In both cycIHC (8-dimensional imaging) and IMC imaging (40-dimensional imaging), Velociraptor accurately identified macrophages using a single search label as input. Notably, Velociraptor worked effectively with both extremely rare and highly abundant cell types and with cell search labels calculated from data and theoretical labels based on literature and expertise. Functional Nano-Color Latex Particles for Inkjet Printing: Synthesis Strategies, Functionalization Techniques, and Advanced Applications.

In modern times, there has been a strong interest in the synthesis of latex particles for futuristic polymer colloid dispersion in the growth of the nanotechnology industry. Functional color latexes (FCL) are synthetic polymer suspension carriers for coloring moieties, with precisely tuned material behavior and properties to suit a particular purpose, attributed to the incorporation of selective functional groups or molecules. Various color latexes have been developed using delicate polymerization techniques such as miniemulsion, microemulsion, suspension, reversible addition-fragmentation chain transfer, and in situ, endowing FCLs with indoor formaldehyde-CO2 removal, antimicrobial activity, self-healing, structural color, anticounterfeiting, rewritable ability, fluorescence, self-cleaning, UV protection, and thermal stability beyond chromatic performance. Over the past few decades, FCL deposition technologies on numerous substrates have enabled widespread applications in smart textiles, flexible electronics, structurally colored coatings, packaging, and functional printing. The present study provides a comprehensive analysis of FCLs' improvement in the borderless application field and their association with synthesis-modification strategies, with attention to their role in intensifying ink properties and empowering digital printing pathways. Future perspectives are concluded by optimizing particle design and determining the appropriate synthesis method to enable feasible production of high-performance latex inks.

Oral and Periodontal Manifestations in Systemic Sclerosis: A Comprehensive Review of Pathophysiology, Clinical Features, and Management Strategies.

Systemic sclerosis is a connective tissue disorder that presents with various orofacial manifestations, which may influence oral health status and complicate dental management. This review sought to identify the oral and periodontal signs linked to systemic sclerosis, examine the specific oral health challenges faced by individuals with this condition, and outline their potential oral healthcare requirements. A review of the available literature was undertaken to evaluate the reported oral and periodontal manifestations associated with systemic sclerosis and to assess their clinical implications. Orofacial manifestations of systemic sclerosis include skin fibrosis, reduced mouth opening, dry mouth, and difficulty swallowing, as well as a potential increased risk of tooth decay, periodontal disease, and oral cancer. These oral and periodontal characteristics, along with their associated complications, improve the understanding and anticipation of oral healthcare requirements for affected patients. Recognition of the oral and periodontal features of systemic sclerosis emphasizes the importance of raising awareness among dental professionals to enhance treatment planning and improve the quality of dental care delivered to this patient population.

Oral Health Status And Oral Health Related Quality of Life in Children With Congenital Heart Disease-A Systematic Review And Meta-Analysis.

The aim of this systematic review was to assess oral health status and oral health related quality (OHRQoL) in children with congenital heart disease (CHD), and to compare the outcomes with children with no systemic diseases. The review primarily focuses on the impact of brushing habits and caries experience on Oral Health related quality of life (OHRQoL) in children with CHD. A comprehensive search of studies published between the timeline of 2004 and 2024 was conducted using PRISMA guidelines. Eleven studies met inclusion criteria for systematic review while ten contributed to the meta-analysis. Data extraction and quality assessment were performed independently by two reviewers using JBI and QATSDD tools.The results showed poor oral health consistently among CHD children, including higher dmft/DMFT scores, compromised oral hygiene, and higher prevalence of enamel defects. The mean difference for caries and oral health indices was 1.52 (dmft/DMFT scores), demonstrating significantly poorer outcomes than systemically healthy counterparts, although neither of these both groups demonstrated optimal oral health status. Behavioural and socioeconomic factors contributed to risk and CHD children showed nearly thrice higher odds of inadequate brushing habits or caries-associated behaviours (OR 3.24, I2 = 0%). OHRQoL was negatively affected in most studies in children with CHD. Children with CHD experience significantly higher compromised oral health and OHRQoL compared to systemically healthy peers. These findings highlight the need for integrated dental preventive care and stronger parental education to improve overall wellbeing.

Improved oral health in head and neck cancer survivors (OHHNC study): a study protocol for a multicentre two-armed randomised controlled trial.

Head and neck cancer comprises a heterogeneous group of tumours affecting anatomical sites of the upper aerodigestive tract. Treatment often involves surgery and either radiotherapy or chemoradiotherapy. These treatments are associated with substantial morbidity and may have long-lasting adverse effects on oral health. Oral complications can negatively affect health-related quality of life, nutritional status, and social and work-related functioning. The aim is to evaluate an oral health care programme designed to prevent or mitigate late oral sequalae, and to assess its impact on health-related quality of life, oral health, work ability, nutritional status, healthcare-related economic costs, and psychological well-being. This is a study protocol for a Swedish multicentre, two-armed, superiority randomised controlled trial that has been ethically approved by the Swedish Ethical Review Authority. Patient inclusion commenced in September 2025, with an intended sample of 300 participants. Adult patients (≥ 18 years) planned for curative-intent treatment for head and neck cancer (oral cavity, oropharynx, larynx stage III and IV, hypopharynx, nasopharynx, salivary gland, or unknown primary) are randomised (1:1) to either an oral health care programme (intervention) or standard of care. severe alcoholism, cognitive impairment, or inability to understand the Swedish language. Participants are followed before the start of cancer treatment and at 4 follow-ups: 6 months after completion of treatment, and 1, 2, and 3 years after the 6-month follow-up. The primary outcome measure is health-related quality of life. Secondary outcomes include quantitative oral health indicators, return to work, nutritional status, healthcare-related economic costs, and patient-reported outcome measures focusing on self-reported oral health and psychological well-being (depression, anxiety and stress), and self-efficacy. Oral health follow-up for head and neck cancer survivors often lacks consistency and uniformity. To date, few oral health care interventions for head and neck cancer survivors have been evaluated within the framework of a randomised controlled trial. The findings have the potential to inform clinical practice and contribute to the development of more integrated, evidence-based oral healthcare strategies within routine follow-up for head and neck cancer survivors, with the overarching goal of improving their oral health and long-term quality of life.
