Profiling Tyrosine Kinase Inhibitor-Specific Signatures of Homocysteine Metabolism and Mineral Homeostasis in Chronic Myeloid Leukemia.

Abdulla Kawakeb N KN, Hade Istikrar M IM, Jumaa Azal Hamoody AH, Hussein Shallal Murad SM et al.

The application of tyrosine kinase inhibitors (TKIs) has revolutionized the management of chronic myeloid leukemia (CML), rendering it a controllable disease. However, long-term administration is associated with adverse metabolic effects, particularly disruptions in homocysteine metabolism and mineral balance. A comprehensive comparison across TKI types remains absent. This study systematically characterized and compared the metabolic signatures elicited by first- and second-generation TKIs Imatinib, Nilotinib, and Dasatinib on homocysteine and mineral parameters in chronic myeloid leukemia. This retrospective cross-sectional study involved 90 stable CML patients treated with TKI therapy for at least six months at Baghdad Oncology Teaching Hospital between March 2024 and April 2025. Patients were divided into three equal groups based on their medication: Imatinib, Nilotinib, or Dasatinib. Fasting blood samples were collected and analyzed for homocysteine, PTH, calcium, potassium, creatinine, vitamin B12, and ALP. Statistical methods, including ANOVA and correlation analyses, were used to compare groups and explore drug-specific effects. The three TKI groups demonstrated distinct metabolic disturbance patterns. Nilotinib was associated with the highest homocysteine levels and showed strong positive correlations with PTH and ALP. Dasatinib caused a moderate elevation in homocysteine and uniquely correlated positively with potassium. Imatinib showed the most favorable homocysteine profile, which was primarily inversely correlated with calcium. Multivariate analysis confirmed that these metabolic profiles effectively distinguished the three TKI groups. The present study demonstrates that Imatinib, Nilotinib, and Dasatinib elicit distinct and discernible metabolic signatures. These results support the adoption of personalized, TKI-specific monitoring approaches in the management of CML, aiming to proactively reduce long-term cardiovascular and skeletal adverse effects and thereby improve alignment between the quality and duration of survival.

Optimal Sheath for Single-Access Percutaneous Coronary Intervention With Impella CP Hemodynamic Support.

Thakker Ravi R, Khalif Adnan A, Xu Ying Y, Bilazarian Seth S et al.

Single-access percutaneous coronary intervention (PCI) with the Impella CP (Abiomed) has increased in prevalence. Despite this, the optimal PCI sheath remains unknown. We characterized the optimal 7F PCI sheath using the single-access technique. The outer diameter measurement of the sheaths was performed using a Keyence 2-axis laser micrometer (Keyence). The force needed to insert the sheath and the torqueability of the guide catheter were assessed using the Mark-10 M5i Advanced Digital Force/Torque Indicator (Mark-10 Corporation) on a simulated single-access setting in a simulated skin pad. The sample size was determined to be 5 sheaths for each test. The BRITE TIP 11 cm (Cordis), BRITE TIP 35 cm (Cordis), and Destination sheath (Terumo) had the largest outer diameters; the Prelude IDeal (Merit Medical) and Terumo Glidesheath (Terumo) had the smallest outer diameters. The Glidesheath was easiest for insertion/removal, followed by the Prelude IDeal and Impella Low Profile (LP) Companion sheath (Abiomed). The Impella LP Companion, Prelude Roadster (Merit Medical), and Destination sheaths allowed for the best torqueability and lowest force to remove the guide catheter. The longer Impella LP Companion sheath did not impact torqueability, but the removal force increased by 50% compared with the shorter Impella LP Companion sheath. The Impella LP Companion sheath demonstrated the best torqueability and least effort in advancing the guide catheters, while the Prelude Roadster and Destination also performed well on these metrics. The Prelude IDeal and Glidesheath were good options for single-access because of their smaller outer diameters and hydrophilic coating and were easiest for insertion/removal through the 14F LP sheath.

A Review of Recombinase-Aided Amplification (RAA) in Veterinary Medicine.

Li Shaomei S, Gao Jieqi J, Miao Yongqiang Y, Chen Qianlin Q et al.

Rapid and accurate pathogen detection is crucial to mitigating the significant economic impacts of infectious animal diseases. Recombinase-aided amplification (RAA) has emerged as a highly promising isothermal diagnostic tool, characterized by its rapid turnaround time, robust amplification efficiency, and single-copy analytical sensitivity. In veterinary diagnostics, RAA has evolved from single-pathogen proof-of-concept assays to become a promising technology for point-of-care scenarios. This review critically evaluates the integration of RAA with real-time fluorescence, CRISPR-Cas systems, lateral flow dipsticks (LFDs), and suspension microarray technologies, highlighting their impact on assay sensitivity and point-of-care (POC) applicability. Despite recent advancements, including encouraging progress in multiplexing and expanded applications in companion animal medicine, inherent challenges such as matrix effects in crude clinical samples, aerosol contamination risks, and commercialization barriers remain. By benchmarking RAA against traditional molecular methods and structurally similar technologies like recombinase polymerase amplification (RPA), this review outlines future trajectories for optimizing RAA platforms, aiming to maximize their utility in routine veterinary surveillance and epidemiological control.

Molecular Dynamics-Guided Triage of Approved Drugs Against Escherichia coli Resistance Targets Nominates Capmatinib, Metergoline and Nilotinib as Candidates for Experimental Testing.

Abdelsayed Mena M, Boulaamane Yassir Y

Virtual screening ranks compounds by scoring functions that correlate with molecular size, and hit lists are rarely benchmarked against compounds of established activity. We screened 2070 approved drugs against four Escherichia coli targets selected by pocket-druggability triage (FabI, LpxC, DHFR and GyrB), with each docking protocol validated by redocking its native ligand (RMSD 0.50-0.69 Å). Per target, the two top-scoring compounds and a reference inhibitor were simulated for 100 ns in triplicate (12 complexes, 3.6 μs). Docking did not recover the references: trimethoprim ranked 762/2068 and novobiocin 576/2069, while top-ranked compounds were large, promiscuous drugs recurring across unrelated targets. Pose triage failed, too: all twelve complexes retained 43-87% of initial contacts, contact-loss time separated targets, not compounds, and MM-GBSA favoured the reference at one target of four. Persistent hydrogen bonding separated the reference from the hits at three of four targets, by 2-9-fold, although the GyrB result rests largely on one replicate. Benchmarking each hit against its reference nominates three candidates for experimental testing: capmatinib at DHFR, metergoline at FabI and nilotinib at LpxC. Capmatinib ranks highest on these computational criteria, with the highest pocket retention (87%) and an MM-GBSA energy with no detectable difference from trimethoprim's at n = 3 (absence of evidence for a difference, not evidence of equivalence). In this small panel, carrying a known active through the same pipeline was therefore more informative than docking score, pose stability or end-point rescoring. The nominations are hypotheses: no experimental validation was performed, and generality would require multiple actives and property-matched decoys per target.

Updates on the Translational Value of Idiopathic Pulmonary Fibrosis Models Across Species: Deeper Insights Through Comparative Pathology.

Gargiulo Sara S, Gramanzini Matteo M, Bonente Denise D, Barone Virginia V

Over the past few decades, advances in comparative pathology have strengthened the potential translational value of animal models of Idiopathic Pulmonary Fibrosis (IPF). The integration of human-relevant diagnostic biomarkers into preclinical studies, such as advanced imaging techniques and standardized histopathological methods, has improved the comparability of experimental findings with human disease. Naturally occurring fibrotic lung diseases in some breeds of companion dogs and cats and induced disease models in laboratory rodents show varying degrees of similarity to human IPF. They represent valuable comparative medicine tools to better understand disease mechanisms and test new therapies, each with advantages and limitations. Critically evaluating their complementarity is essential for gaining reliable insights into disease processes through preclinical animal studies and for developing effective treatments. This comprehensive review aims to update current knowledge of these animal models of IPF, focusing on comparative pathology, imaging, and histological features and evaluating their respective strengths, limitations, and translational relevance within the conceptual framework of "One Health, One Medicine, One Pathology". By exploiting convergent pathophysiological processes across species, this interdisciplinary collaborative paradigm aims to promote the effective use of model organisms in biomedical research and foster mutual benefits for both humans and animals.

Comparative Veterinary Psychopharmacology in Dogs and Cats: Neurotransmitter Targets, Translational Boundaries, and Species-Specific Pharmacokinetics.

Kim Iljin I, Yun Jang-Hyuk JH

Behavioral disorders in companion animals are increasingly being managed with pharmacotherapy. However, prescription practices often rely on extrapolation from human psychiatry without considering species-specific phenotypes, diagnostic frameworks, or pharmacokinetic variability. A structured translational approach is required to align mechanistic plausibility with clinical evidence and interspecies constraints. This review consolidates the current information on the pharmacological modulation of neurotransmitter systems in canine and feline behavioral disorders using a comparative framework. Neurotransmitter circuits are mapped to relevant phenotypes. Controlled clinical evidence supports the use of selected serotonergic agents for specific chronic behavioral indications in dogs, whereas evidence in cats remains scarce and indication-specific. Controlled veterinary evidence is also available for selected agents used in acute or situational behavioral indications and for selegiline in canine cognitive dysfunction. Contrastingly, the broader neurotransmitter-phenotype relationships and emerging pharmacological targets discussed in this review are supported by mechanistic or translational evidence and should not be interpreted as established clinical indications. Cross-species translation is most defensible at the level of conserved receptors and transporters, whereas direct transfer of human diagnostic categories and subjective symptoms is limited in veterinary patients. Species- and drug-specific pharmacokinetic differences, along with limited feline data, necessitate careful dose initiation, titration, and monitoring to mitigate adverse effects and drug interactions. This review provides a structured framework for interpreting psychopharmacological evidence in dogs and cats by integrating pharmacological mechanisms, the design and scope of available clinical studies, translational boundaries, and species- and drug-specific pharmacokinetic considerations. Future research could benefit from standardized outcome measures, expanded feline trials, and pragmatic real-world designs.
